螺旋藻中性多糖硫酸化及性质研究

摘    要:采用氯磺酸-吡啶法对螺旋藻中性多糖进行硫酸化修饰;玫瑰酸钠法测定修饰前后的多糖SO4^2-含量分别为0.17%、20.0%;取代度Ds=0.429;利用琼脂糖凝胶电泳对多糖硫酸酯修饰物进行研究。结果表明多糖硫酸酯修饰物的琼脂糖凝胶电泳呈一均一斑点.与其他几种硫酸化多糖相比迁移率有较大差异;红外光谱法确定硫酸酯键在1270~1200cm^-1.1000~800cm^-1处存在强的特征吸收峰;HPLC显示出一个主要的单峰,说明此制备方法可行。

Abstract:A neutral polysaccharide from Spirulina Platensis (NPSP) as sulfated by chlorosulfonic acid pyridine method. With the method of sodium rhodizonate, the content of sulfate group of NPSP and NPSP sulfate was 0.17% and 20.0%. The average of substitution D s was 0.492. Using agarose gel electrophoresis (AGE), the electrophoretic behaviors of NPSP sulfate had one spot and compared with other kinds of sulfate polysaccharides, its migration ratio was differen. Some strong characteristic absorption peaks of ester sulphate bonds were observed at 1 270 ~ 1 200 cm 1 and 1 000 ~800 cm 1 respectively by FTIR. A major single peak in HPLC showed the method was successful.
