嗅鞘细胞移植治疗EAE的研究

目的 探讨嗅鞘细胞（OECs）移植对实验性免疫性脑脊髓炎（EAE）的治疗 作用。方法 用新生wistar大鼠嗅球培养OECs。OECs移植组将OECs悬液注入EAE大鼠的侧脑室，假手术组注入等量生理盐水；观察两组大鼠运动功能评分、脑组织病理学变化及热休克蛋白70（HSP70）的表达。结果 OECs移植组运动功能评分显著优于假手术组（P〈0．05）；OECs移植组髓鞘修复明显优于假手术组；OECs移植组在各时间点上HSP70的表达均明显高于假手术组（P〈0．05）。结论 OECs移植可以促进EAE鼠的髓鞘修复，改善其运动功能，同时可使其脑组织HSP70大量表达，对损伤后的神经细胞有保护作用。

Observation of OECs transplantation for experimental allergic encephalomyelitis

Objective:To develop a method for olfactory ensheathing cells(OECs)culture and to evaluate the therapeutic effect of OECs transplantation on experimental allergic encephalomyelitis(EAE).Methods: OECs were cultured in the olfactory bulb of newborn wistar rat.OECs suspension was infused into the lateral cerebral ventriculus of EAE rats,and the saline with the same quantity was infused in pseudo operation group.Afterwards,sport function grade histo pathological changes and HSP70 expression in cerebral tissues were observed.Results: The sport function grade and myeline repair of OECs transplantation group was significantly superior to those of pseudo operation group(P< 0.05),and the HSP70 expression of OECs group was markedly higher than that of pseudo operation group at each observation time(P<0.05).Conclusion: OECs transplantation contributes to myeline repair of EAE,improvement of sport function of experimental rats and HSP70 expression,and it plays a protective role on injured neurons.
