大鼠胶原诱导关节炎模型滑膜肽基精氨酸脱亚胺酶4表达

目的探讨肽基精氨酸脱亚胺酶4(peptidylarginine deiminase 4,PADI4)在大鼠胶原诱导关节炎模型的表达。方法2012年7月至2013年5月中国医科大学附属盛京医院实验中心将关节炎模型建立成功且关节炎指数大于2分的18只大鼠随机分为胶原诱导关节炎(collagen-induced arthritis,CIA)3周模型组、4周模型组及6周模型组各6只,另有6只大鼠为对照组。分别予以生理盐水1 mld(对照组)、0.5 mgml鸡Il型胶原(CIA模型组)皮内注射,并于免疫后3、4、6周按组别处死。比较各组大鼠体重、关节炎指数、足趾容积及PADI4表达的变化情况。结果 CIA模型组关节炎发展迅速,关节炎指数、足趾容积相对于正常对照组明显增高,PADI4表达亦增多,体重减轻。结论 PADI4与大鼠胶原诱导关节炎模型有明显的相关性。

Expression of Peptidylarginine Deiminase 4 in the Synovium of Collagen Induced Arthritis Rat

Objective To study Peptidylarginine deiminase 4 （PADI4） and its expression in Collagen induced arthritis rats （CIA）. Methods From July 2012 to May 2013, the model of arthritis was suceessfully built in the Experimental Center, Shengjing Hospital Affiliated to China Medical University and those 18 rats with score higher than 2 were randomized into three collagen induced arthritis （CIA） groups, labeled as 3 weeks executed group, 4 weeks executed group and 6 weeks executed grou Prespectively, with 6 in each. Another 6 rats were recruited as the normal controls. Normal saline 1 ml （control group）, 0. 5 mg/mL chicken type 1I collagen （CIA model group） were intra-cutaneously injected everyday before the rats were sacrificed alter immunization in the 3＇h, 4~h and 6th week respectively according to the designated group. The body weight, arthritis index, the volume of toe, PADI4 expression levels were compared between groups. Results The arthritis developed quickly in CIA model group, the arthritis index and the volume of toe increased obviously, and the body weight decreased, with elevated expression of PADI4. Conclusion PADI4 is obviously correlated with CIA.
