| 摘 要： | 目的 探讨大鼠肾移植手术前后趋化因子CXCL9及其受体CXCR3 mRNA的动态变化在术后急性排斥早期诊断的作用.方法 以BN和LEW大鼠作为供体,LEW大鼠作为受体,建立大鼠同种异体肾移植模型.采用ELISA法和逆转录聚合酶链反应(RT-PCR)分别动态检测大鼠术前后血清CXCL9和外周血淋巴细胞CXCR3 mRNA的变化.同时检测血清肌酐水平.结果 实验组大鼠血清CXCL9、外周血淋巴细胞CXCR3 mRNA及血清肌酐表达均明显升高,与对照组比较差异有统计学意义(P<0.01).术前实验组与对照组血清CXCL9和外周血淋巴细胞CXCR3 mRNA表达比较无统计学差异(P>0.05),术后第1、3、5、7天实验组均高于对照组,差异有统计学意义(P<0.05).术前及术后第1天实验组和对照组血清肌酐表达比较无统计学差异,术后第3、5、7天实验组均高于对照组,差异有统计学意义(P<0.05).结论 血清趋化冈子CXCL9和外周血淋巴细胞CXCR3 mRNA的表达与排斥反应密切相关,可以作为早期诊断急性排斥反应的辅助敏感指标.
