组织胺对大鼠DRG神经元ATP-激活电流的抑制作用

摘    要:目的:探讨组织胺对大鼠DRG神经元ATP-激活电流的调制作用.方法:采用全细胞膜片钳技术,在新鲜分离的大鼠背根神经节细胞上进行.结果:实验观察到组织胺在DRG神经元可引起内向电流,并有明显的浓度依赖性.在被检测的DRG神经元中,85%(30/35)的细胞对ATP敏感,可引起一浓度依赖性的去敏感的内向电流.预加10-8、10-7、10-6、10-5、10-4mol/L组织胺后,对ATP-激活电流的抑制分别是:(12.50±3.2%(n=5)、(24.49±3.5%(n=5)、(35.18±4.5%(n=6)、(32.62±5.8%(n=8)、(23.53±4.2%(n=5),呈浓度依赖性.结论:组织胺对大鼠DRG神经元ATP-激活电流有明显的抑制作用.

Abstract:Objective:To investigate the regulation of Histamine on ATP-activated current in rat dorsal root ganglion(DRG) neurons.Methods:The effect of Histamine on ATP-activated current was studied using the whole-cell patch-clamp technique in fresh isolated dorsal root ganglion neurons.Results:Histamine can activate detectable inward current in 55%(10/18) DRG neurons and Histamine-activated current was dependent upon concentration.Most of cells tested was sen-sitive to ATP,90%(45/50)cells can activate a desensitized inward current in a concentration-d ependentmanner.After pre-applicationHistamine,Histaminedecrease theamplitude of ATP-activated current in a concentration-dependent manner over the concentration range of 10-8,10-7,10-6,10-5,10-4 mol/L,with a(12.50±3.2 %(n=5),(24.49±3.5 %(n=5),(35.18±4.5 %(n=6),(32.62±5.8 %(n=8),(23.53±4.2 %(n=5) inhibition for current activated by ATP.Conclusion: The results suggest that Histamine can regulate ATP-activated current.
