成人胰岛来源的Nestin阳性前体细胞的体外培养及其诱导分化

目的 探讨成人胰岛来源的Nestin阳性前体细胞的体外培养、扩增及诱导分化为具有分泌胰岛素功能的B细胞的条件。方法 首先通过胶原酶消化的方法从成人胰腺组织中分离纯化出胰岛，在含有bFGF及EGF的增殖培养基中大量扩增获得Nestin阳性前体细胞，最后在含有HGF、activin-A、exendin-4等生长因子的培养基中诱导该前体细胞。结果 来源于成人胰岛的Nestin阳性前体细胞在体外可以大量扩增并表达ABCG2，而胰岛素不表达。在分化培养基中能分化为对高糖刺激反应的胰岛B细胞，胰岛素呈现阳性。结论 来源于成人胰岛的Nestin阳性前体细胞能够为今后糖尿病的细胞移植治疗提供大量有功能的B细胞，能够解决目前胰岛移植中供体来源不足的问题。

Cultivation and Inducing Differentiation of Nestin Postive Precursor Cell Origined from Adult Pancreatic Island

Objective To explore the conditions under which nestin-positive precursor cells can be isolated,cultured, proliferated and induced into B cells.Methods Islets were isolated by collagenase digestion method,and nestin-positive cells can be proliferated in media containing bFGF and EGF.Results The nestin-positive cells origined from adult might proliferated and expressed ABCG2 in vitro,but insulin didn't expressed.In addition,these cells also could be induced into B cells that could response to glucose-challenged.Conclusion Nestin-positive islet-derived progenitor cells can provide lots of B cells for future islet application and solve the problem with short of donor organ.
