雷公藤内酯醇对卵巢癌细胞株A2780作用的实验研究

摘    要:目的:观察雷公藤内酯醇对卵巢癌细胞株的A2780的抑制作用,以及与顺铂联合用药的协同作用,并与顺铂单独作用相比较,评价其作为抗肿瘤药物的价值。方法:MTT检测不同的用药方式对卵巢癌细胞A2780增殖的抑制作用;形态学观察经雷公藤内酯醇作用后细胞的生长情况;流式细胞仪检测雷公藤内酯醇作用后细胞的凋亡情况。结果:1)雷公藤内酯醇对卵巢癌细胞A2780的增殖有明显抑制作用,呈量-效、时-效关系(P<0.05)。其在较小的浓度下即可产生很好的抑制效果(5 mg/L时,抑制率80%;0.5 mg/L时,抑制率60%;(0.005 mg/L)时,抑制率40%)。2)雷公藤内酯醇与顺铂相比较,在浓度<5 mg/L雷公藤内酯醇队卵巢癌细胞A2780的抑制作用优于顺铂(P<0.05),但在大于此浓度下则两者无明显差异(P>0.05);在24、48、72 h作用点时,雷公藤内酯醇IC50均小于顺铂IC50。3)在低浓度下,雷公藤与顺铂两者联合用药后的抑制率大于顺铂单用药抑制率,而小于雷公藤内酯醇单用药抑制率,其抑制率受作用时间影响更大,随作用时间的延长其抑制率明显增高,尤其在作用48 h以后,抑制率增加更为明显(P<0.05)。4)流式细胞检测经雷公藤内酯醇作用后A2780细胞有凋亡(P<0.05)。倒置相差显微镜下观察经雷公藤内酯醇作用后细胞生长明显受到抑制。结论:雷公藤内酯醇对卵巢癌细胞株A2780有明显的抑制作用,有量-效、时-效关系;在较低的浓度下,其抑制率明显高于顺铂;雷公藤内酯醇与顺铂联合用药的作用,随时间的延长抑制率增大。雷公藤内酯醇抑制肿瘤的作用可能是通过诱导细胞凋亡实现的。

Abstract:Objective:To evaluate the value and significance of triptolide as an antitumor drug.The inhibitive effect of triptolide and the synergistic effect of triptolide combined with cisplatin and comparison of the effect between triptolide and cisplatin on ovarian cancer cell line A2780 are investigated.Methods:Methyl thiazolyl tetrazolium(MTT)assay was used to examined the inhibitive effect of triptolide on ovarian cancer cells' proliferation in vitro,the cells' proliferation affected by triptolide was observed by microscope,and the cells' apoptosis after treatment with triptolide was observed by FCM.Results:1)Triptolide had significant effect on ovarian cancer cell line A2780 in a dose-dependent and time-dependent manner(P<0.05);At lower concentration(5 mg/L,0.5 mg/L,0.005 mg/L),the inhibitive rate was 80%,60% and 40% respectively.2)Comparing with cisplatin,triptolide had more significant inhibitive effect on A2780 at low concentration(<5 mg/L)(P<0.05),but at high concentration(>5 mg/L)their inhibitive effects were not statistically significantly different(P>0.05);and the IC50 of triptolide was less than that of cisplatin in 24 h,48 h,and 72 h after treatment.3)At low concentration(<5 mg/L),the inhibitive rate of combination triptolide with cisplatin was higher than that of cisplatin alone,but lower than that of triptolide alone.And the inhibitive rates increased with the prolongation of treated time,especially when the treated time was longer than 48 h.4)Apoptosis existed in A2780 after treatment with triptolide(P<0.05).Conclusion:Triptolide has significant suppressive effect on ovarian cancer cell line A2780 in a dose-dependent and time-dependent manner(P<0.05),and it works even in a low concentration(<5 mg/L),At lower concentration,the inhibitive rate of triptolide is higher than that of cisplatin.The inhibitive rate of combined triptolide and cisplatin increases with the prolongation of affected time.The anti-tumor effect of triptolide may be achieved by inducing cells' apoptosis.
