帽儿山地区异色瓢虫遗传多样性的ISSR分析

摘    要:利用ISSR技术对东北帽儿山地区13种不同色斑型异色瓢虫进行基因组多态性分析,从40条引物中筛选出13条用于异色瓢虫的ISSR扩增。共扩增出127条带,其中多态性条带120条,多态性百分率为94.49%。根据ISSR扩增结果,利用NTSYSpc2.10e软件进行Jaccard相似系数分析,13种不同色斑型异色瓢虫的遗传相似系数为0.36~0.60,平均相似系数为0.51。通过UPGMA聚类分析,在遗传相似系数0.50处,13份瓢虫材料可分为2大类群。主成分分析结果表明,显明变种、四斑变型和二窗无脊3种斑型瓢虫是异色瓢虫遗传变异的主线。ISSR分析表明,异色瓢虫的遗传多样性较为丰富。笔者还根据聚类结果对瓢虫鞘翅底色变异的可能因素进行了分析。

Abstract:The genomic polymorphism of 13 species of Harmonia axyridis(Pallas)with different splash types from Maoershan area was analysed using inter-simple sequence repeat(ISSR)markers.Thirteen among 40 ISSR primers were chosen for ISSR amplification.A total of 127 bands were generated,of which 120 bands were polymorphic bands(94.49%).Jaccard similarity coefficient analysed by NTSYSpc2.10e software showed that the genetic similarity coefficient was from 0.36 to 0.60 and the average genetic similarity coefficient was 0.51 according to the result of ISSR amplification.The clustering dendrogram was constructed by UPGMA method,and thirteen samples were divided into four major groups at a similarity coefficient of 0.50.Principal component analysis indicated that three splash types(H.axyridis var.spectabilis Fald,H.axyridis ab.34-quattuorsignata(Hem.)Yuan.,H.axyridis.ab.succinea Hope)were the masterstroke of heredity aberrance in H.axyridis(Pallas).ISSR analysis exhibits a high genetic diversity of H.axyridis(Pallas).Moreover,the variance of the vagina wing impression in H.axyridis was also analysed by the clustering result.
