Abstract:Uterine corpus endometrial carcinoma(UCEC) is one of the most dangerous cancers for women’s health, but the mechanism of its development is not fully understood. Aberrant expression of genes plays an important role in the process of cellular carcinogenesis. This study intends to use bioinformatics to analyze the network regulation of abnormally expressed genes in UCEC to provide theoretical basis for the mechanism study and prognostic treatment of UCEC. Firstly, the differentially expressed RNA molecules were screened by "limma" package. Then, the lncRNA-miRNA-mRNA ceRNA network was constructed by using the "GDCRNATools" package in the R software. Finally, we used The Database for Annotation, Visualization and Integrated Discovery(DAVID), the "survival" package in the R software, the Molecular COmplex Detection(MCODE) plug-in, and the StarBase database for comprehensive analysis of the RNA molecules in the ceRNA network. The results showed that 1 319 mRNAs, 68 lncRNAs and 100 miRNAs were identified as differentially expressed RNA molecules in UCEC. At the same time, using these differentially expressed RNA molecules in UCEC, the ceRNA network was successfully constructed. The results showed that HCG11, LINC00958, LINC00667, MAGI2-AS3 and AC093010.3 had the function of ceRNA. Through further analysis of the network members, it was found that the mRNA molecules in the network were enriched in the biological processes of endothelial cell proliferation, cytoskeleton and negative regulation of zinc ion binding. Survival analysis showed that 19 differentially expressed RNA molecules such as hsa-miR-449a, LINC00958, PKIA, and DPYSL2 were significantly associated with the prognosis of patients with UCEC. Finally, after screening by the MCODE plug-in, this study yielded a subnetwork whose members were LINC00667, hsa-miR-449a, hsa-miR-34a-5p and RECK. The results of this study suggested that there was differential expression of lncRNA, miRNA and mRNA in the development of UCEC. These RNA molecules formed the regulation of the ceRNA network pattern and some of these key molecules were associated with patient prognosis, which will provide a theoretical basis for studying and understanding the pathogenesis and prognosis of UCEC.
