摘    要:目的 寻找慢性砷中毒的最佳生物标志物,建立砷中毒的快速检测及标志物的形态分析方法。方法 以1例砷中毒患者的血、尿、头发为主要样本来源,采用电感耦合等离子体质谱测定总砷、液相色谱原子荧光光谱仪联用技术测定患者头发中的砷形态。结果 患者就医初期血砷(31.5±2.6)μg/L]、尿砷(126.0±3.4)μg/L]、发砷(49.6±0.7)μg/g]均高于参照组及文献报道正常水平,且发砷异常最为明显;动态监测结果显示,治疗后发砷含量下降最快。所建立的发砷含量和发砷形态分析方法的线性范围分别为1.00~50.00、2.50~50.00μg/L,相关系数r>0.999 4,回收率为85.4%~98.1%。结论 所建立的以发砷为标志物的砷中毒及形态分析方法,准确、快速,可用于慢性砷中毒的快速测定,并可通过动态监测为砷中毒的治疗提供指导。

Abstract:Objective] To search for the best biomarkers of chronic arsenic poisoning and establish a rapid detection and speciation analysis method for arsenic poisoning.Methods]The blood,urine and hair of a case of chronic arsenic poisoning were collected as main samples. The arsenic content in these samples were detected using inductively coupled plasma mass spectrometry and the arsenic species in these hair were analyzed by atomic fluorescence spectrometry combined with high performance liquid chromatography.Results] At the initial stage of clinic treatment,the content of arsenic in patient’s blood(31.5±2.6)μg/L],urine(126.0±3.4)μg/L] and hair(49.6±0.7)μg/g] were higher than those in the reference group and exceeded the normal level reported in research. Furthermore,the arsenic content in patient’s hair was obviously abnormal. The results of dynamic monitoring showed that the arsenic content in hair decreased mostly after treatment. The linear range of arsenic content and arsenic speciation analysis in hair were1.00-50.00 and 2.50-50.00 μg/L respectively. The correlation coefficients were more than 0.999 4 and the recoveries were within 85.4%-98.1%.Conclusion] The method for arsenic poisoning and the arsenic species analysis based on arsenic content in hair is accurate,rapid,and could be used for rapid determination of chronic arsenic poisoning. Furthermore,it could provide guidance for the treatment of arsenic poisoning through dynamic monitoring.
