摘    要:目的 研究负载金黄色葡萄球菌肠毒素B(staphylococcalenterotoxinB ,SEB)和金黄色葡萄球菌肠毒素C(staphylococcalenterotoxinC ,SEC)的DC在体外对T细胞的活化能力及其对肝细胞癌HcaF细胞的杀伤作用。 方法 以SEB/SEC负载DC刺激T细胞 ,流式细胞仪 (flowcytometer ,FCM )检测T细胞受刺激后其早期活化标志CD69表达情况和IL 2水平 ,采用MTT法检测T细胞增殖水平和其对HcaF细胞的杀伤效应。结果 SEB/SEC联合DC在体外能明显激活T细胞 ,以浓度为 10 0ng/ml时作用最强 ;SEB/SEC联合DC可以使T细胞活化、增殖并大量分泌IL 2 ,对HcaF细胞的杀伤率高达 83 .2± 12 .8%。以SE作为刺激物负载的DC较以Tag作为刺激物负载的DC有更强的活化T细胞的能力 ;结论 根据以上结果 ,可以认为SEB/SEC联合DC具有很强的刺激T细胞活化的能力 ;其活化的T细胞具有很强的杀瘤作用 ,在肿瘤免疫治疗方面展现出良好的潜在应用前景。

Abstract:Objective To evaluate the effect of staphylococcal enterotoxin B (SEB) or staphylococcal enterotoxin C (SEC) combining with dendritic cells (DC) on T cells and the anti-HcaF tumor cytotoxicity of activated T cells in vitro. Methods The CD69 expression on T cells and IL-2 production level were verified using the flow cytometer. The proliferation and cytotoxicity of T cells were detected by MTT assay. Results SEB/SEC combining with DC could significantly activate T cells in vitro and the most powerful concentration was 100 ng/ml. Activated T cells proliferated and produced high level of IL-2. Activated T cells and its supernant showed significant effect of killing HcaF cells. SEB/SEC-loaded DC was a more powerful stimulus for T cells than tumor antigens-loaded DC. Conclusions SEB/SEC combining with DC is a stronger stimulus for activating T cells to produce a forceful anti-HcaF tumor cytotoxicity than tumor antigen-loaded DC used at present. It may be applied as a vigorous weapon to clinical tumor immunotherapy.
