摘    要:利用微生物酶法转化白头翁总皂苷,结合硅胶柱层析法和结晶法对酶产物进行分离,制备高活性的稀有白头翁皂苷A3。将1 000g中药白头翁经乙醇浸提3次,再经脱糖、脱色处理后共得总皂苷98.6g,得率为9.86%。采用微生物Absidiasp.P00r发酵产粗酶液,对50g总皂苷进行酶反应,酶解产物经脱糖、脱色处理后,得到含白头翁皂苷A3的粗产物34.2g。通过硅胶柱层析和结晶法对产物进行分离纯化,制备得到白头翁皂苷A3单体1.56g,得率为4.56%,HPLC检测其纯度为95.4%。

Preparation of anemoside A3 by biotransformation
Abstract:The minor anemoside A3was transformed enzymatically from total pulsatilla saponin,and purified by silica-gel column chromatography and crystallization.The dried root of Pulsatilla chinensis(Bge.)Regel(1 000g)was extracted with alcohol for three times,and obtained with a yield of 9.86%after decoloration and desugar.The crude product of 34.2g was obtained from 50g total saponin by transformation with enzyme from Absidia sp.P00rafter decoloration and desugar. Finally,1.56g pure anemoside A3with a yield of 4.56% was obtained after separation by silica-gel column chromatography and purification by crystallization.The purity of anemoside A3was above 95.4% based on HPLC analysis.
