摘    要:正苹果褪绿叶斑病毒(Apple chlorotic leaf spot virus,ACLSV)属于乙型线形病毒科(Betaflexiviridae)纤毛病毒属(Trichovirus),基因组为正义单链RNA,含有约7 500个核苷酸,编码3个部分重叠的开放阅读框。目前从Gen Bank数据库可以检索到19个ACLSV分离物的基因组序列。该病毒侵染仁果与核果,如苹果和桃,在苹果栽培种上不引起症状,在特定指示植物上引起症状,如俄罗斯苹

Abstract:In the present work, we studied the genomic and biological properties of Apple chlorotic leaf spot virus (ACLSV) isolate XC-HF. The leaves of Malus domestica cv. Hanfu collected from Xingcheng, Lianoning province were used as plant material. The samples were subjected to RT-PCR and RACE techniques to amplify the complete genome of ACLSV XC-HF, followed by sequencing. The genome of ACLSV XC-HF consists of 7 557 nucleotides, encoding three overlapping open reading frames (ORFs) and shared nucleotide identities of 68.9%-84.4% with the other 19 ACLSV isolates published in GenBank. Phylogenetic analysis based on the complete genome sequences indicated that ACLSV XC-HF was located in the group JB. Recombination analysis suggested a recombinant event at 1-338 nts of ACLSV XC-HF. The ACLSV XC-HF was inoculated to Chenopodium amaranticolor by sap inoculation and to Malus sylvestris cv. R12740-7A by double budding, respectively, and induced symptoms of chlorosis and slight leaf roll on infected plants.
