使用免疫印迹试验筛选与不孕相关的精子抗原研究

摘    要:采用精子抗体阳性的免疫性不育病人血清,并通过免疫印迹试验,筛选与不孕相关的精子抗原,这对于避孕疫苗研制和免疫性不孕诊治均具有十分重要的意义。本研究结果显示,在ELISA试验测定精子抗体阳性的14例中,免疫印迹试验阳性11例(78.6%)。其中8例(72.7%)能识别92kD,6例(54.5%)能识别58 kD,3例(27.3%)能识别72 kD,2例(18.2%)能识别205、88和56 kD,只有1例(9.1%)可识别49和46kD的精子膜抗原。其中92、72、58kD精子膜抗原被识别的频度较高,提示这些抗原分子表位所诱发的抗体与不孕相关较为密切。

Abstract:It is very important to identify sperm antigens for immunoinfertility and immunocontraceptive vaccine research.The sperm antigens associated with infertility were selected by using immunoblotting assay fram human sera with sperm antibody. The results showed that in 14 cases with positive sperm antibody detected by enzyme linked immunosorbent assay (ELISA).11 cases(78. 6%)were positive by immunoblot analysis. Among these 11 cases with positive results detected by immunoblot assay. 8 cases (72. 7%)could recognize 92 kD antigen of human sperm membrane,6 cases(54. 5%)could bind with 58 kD antigen, 3 cases (27. 3%) could react with 72 kD antigen,2 cases could recognize 205,88 and 56 kD antigens,only one case could react with the sperm antigens with molecular weight va-lues of 49 and 46 kD. These results indicated that in immunoblot assay,the incidences of 92,72,58 kD sperm antigens recognized by immunoinfertile sera with positive sperm antibodies were higher than those of other antigens. It is suggested that these antibodies induced by epitopes of 92,72 and 58 kD antigens of human sperm membrane may associate with immunoinfertility. Therefore, further studies on the 92,72 and 58 kD sperm antigens should be performed for developing sperm immunocon-traceptive vaccine.
