马铃薯茎尖组织培养方法优化研究

摘    要:提高马铃薯茎尖组织培养的成苗率和脱毒率。以克新1号为材料,研究了不同茎尖大小、不同浓度和配比的激素对马铃薯茎尖诱导的效果。结果表明:茎尖越小,脱毒率越高,但成活率和成苗率越低,以叶原基数为2的茎尖脱毒效果最好。通过对6种培养基的对比,筛选出MS+6-BA 0.5 mg/L+NAA 0.1 mg/L+GA 0.1 mg/L为茎尖诱导分化成苗的最优培养基。

Study on Optimizing Method of Potato Stem tip Tissue Culture

Abstract:To improve the potato stem tip tissue culture seedling rate and detoxication rate. Using the new 1 as material, study the effect of different size of shoot tip, concentrations and proportions of hormone on potato stem tip induction. The re- suits show that the smaller stem apex, virus -free rate is higher, but the survival rate and seedling rate is lower, with Ye Yuan base 2 stem apex detoxification effect best. Through the 6 medium contrast, screening out MS + 6 - BA 0. 5 mg/L + NAA 0. 1 mg/L + GA 0. 1 mg/L stem tip induction seedling differentiation of optimal culture medium.
