宫颈癌中Notch1和PTEN的表达及相关性研究

目的:检测Notch1和PTEN在正常宫颈组织、宫颈上皮内瘤样病变(CIN)和宫颈癌组织中表达情况,并探讨两者在宫颈癌中表达的临床意义。方法:采用免疫组织化学染色方法(S-P法)检测50例宫颈癌、31例CIN和20例正常宫颈组织中Notch1和PTEN的表达情况。结果:Notch1和PTEN表达定位于细胞浆和细胞核,其中Notch1在宫颈癌中阳性表达率为84.%,明显高于两者在CIN组和正常宫颈组织的表达率,PTEN在宫颈癌的阳性率为32%,低于CIN组和正常宫颈组织的表达率。两者在宫颈癌的表达情况与有无伴有淋巴结转移、临床分期及病理分期有关,其差异有统计学意义(x2=7.948,x2=5.082,x2=7.317;P<0.05);在宫颈癌组织中Notch1和PTEN两者之间比较表达呈负相关(r=-0.402,P<0.05)。结论:Notch1与PTEN的表达失调可能与宫颈癌的发生、发展相关,可做为评价宫颈癌预后的指标。

Expression and significance of Notchl and PTEN in cervical carcinoma

[Abstract] objective： To determine the expression of Notch-1 and PTEN in the normal cervical tissue, CIN and cervical carcinoma and discuss the clinical significance of both in cervical cancer. Methods： The expression of Notch1 and PTEN in 50 cases of cervical carcinoma, 31 cases of CIN and 20 cases of normal endometrial tissue was detected by immunohistochemistry S-P Results： T Notchl and PTEN expression located in the cytoplasm and nucleus. The expression rate of Notch1 in cervical cancer was 84.00%（42/50）, which was obviously higher than that of the both in CIN group and normal cervical tissue（P〈0.05）. The positive expression rate of PTEN in cervical cancer was 32.00%（16/50）, which was markedly lower than that in the CIN group and normal cervical tissue expression rate（P〈0.05）. Both of them were associated with lymph node metastasis and clinical staging and pathologic staging（P〈0.05）. In cervical cancer tissues, the expression of Notchl was negatively correlated with that of PTEN（P〈0.05）. Conclusion： The expression of Notchl and PTEN may be related with cervical cancer occurrence and development. Both of them can be used as the indexes for prognosis of cervical cancer.
