黄芪甲苷对自发性高血压大鼠血管内皮功能的保护作用

摘    要:【目的】探讨黄芪甲苷对自发性高血压大鼠(SHR)血管内皮功能的保护作用。【方法150只SHR大鼠随机分为模型组,黄芪甲苷低、中、高剂量组5,10,20mg/(kg·d)],阳性对照组(10mg/kg卡托普利),另外选取正常血压的Witstar-Kyoto大鼠10只作为正常对照组。采用BP-6动物无创血压测试仪检测各组大鼠治疗前血压,再连续给药8周,1次/日,在第2、4、6、8周测量各组大鼠血压,第8周处死大鼠,主动脉取血,检测各组血清中一氧化氮(NO),内皮素-1(ET-1),血管紧张素Ⅱ(AngⅡ),血栓素A2(TXA2),超氧化物歧化酶(SOD),丙二醛(MDA)水平并比较。【结果】与模型组比较,黄芪甲苷低、中、高剂量组血压、ET-1、AngⅡ、TAX2及MDA含量依次降低(P〈0.05),NO及SOD含量依次升高(P〈0.05)。【结论】黄芪甲苷能降低SHR血压,改善血管内皮功能,可能与调节血管收缩和舒张因子表达及氧化应激水平有关。

Protective effects of Astragaloside on the Function of Vascular Endothelium in Spontaneous Hypertension Rats
Abstract:Objective]To explore protective effects of astragaloside on the function of vascular endothelium in spontaneous hypertension rats (SHR) .Methods]Fifty SHRs were randomly divided into the model group ,the low ,medium and high‐ as‐tragaloside‐dose groups 5 ,10 ,20 mg/( kg .d)] ,the positive control group (10 mg/kg ,captopril ) .In addition ,10 Witstar‐Kyoto rats with normal blood pressure were selected as normal control group .The blood pressure of rats were detected before treatment by BP‐6 noninvasive blood pressure detector followed by medicine administration once a day for 8 weeks .Blood pres‐sure were measured in second ,forth ,sixth and eighth week regularly .At eighth week ,the rats were killed and the blood was taken from the aorta .The levels of nitric oxide (NO ) ,endothelin‐1 (ET‐1 ) ,angiotensin Ⅱ (Ang Ⅱ ) , thromboxane‐A2 (TXA2) ,superoxide dismutase (SOD) and malondialdehyde (MDA) of serum in each group were detected respectively .Re‐sults]Compared with the model group ,the blood pressure ,ET‐1 ,Ang Ⅱ ,TAX2 and MDA contents of the low ,medium and high astragaloside ‐dose groups were decreased in turn (P < 0 .05) ,and the contents of NO and SOD were increased in turn( P< 0 .05) .Conclusion] Astragaloside could decrease the blood pressure and improve vascular endothelial function in SHR ,which may be related to the regulation of vascular contraction ,the expression of relaxation factor and to the level of oxidative stress .
