摘    要:目的 探讨五花血藤提取物(sargentodoxa cuneata extracts,SCE)对大鼠视网膜缺血-再灌注损伤(RIRI)中凋亡相关因子Bcl-2、Bax和Caspase-3蛋白表达的影响。方法 选取健康清洁级成年SD大鼠90只,随机分成对照组、RIRI组与SCE组,每组30只。于RIRI后6 h、12 h、24 h、48 h与72 h时,每组分别随机处死6只大鼠进行相关实验。通过Nissl染色观察各组大鼠视网膜神经细胞形态学的变化,免疫组织化学法检测各组大鼠视网膜内各细胞层凋亡相关因子Bcl-2、Bax和Caspase-3的表达。结果 对照组视网膜神经节细胞内Nissl小体丰富,染色较深,呈块状分布;RIRI组Nissl小体较对照组少,染色浅且分布不均;SCE组染色情况均较RIRI组有所改善。Bcl-2蛋白在对照组中几乎不表达,在24 h RIRI组中其阳性表达开始增强,48 h时达到高峰,在SCE组各个时间点上Bcl-2蛋白阳性表达均多于RIRI组,差异均有统计学意义(均为P<0.05)。Bax蛋白在对照组中略有表达,在12 h RIRI组中其阳性表达持续增多,24 h时达到高峰,在SCE组各个时间点上Bax蛋白阳性表达均低于RIRI组,差异均有统计学意义(均为P<0.05)。Caspase-3蛋白在对照组中几乎不表达,在6 h RIRI组中表达开始升高,12 h时达到高峰,在SCE组各个时间点上Caspase-3蛋白阳性表达均低于RIRI组,差异均有统计学意义(均为P<0.05)。结论 SCE可能通过上调Bcl-2蛋白表达的同时下调Bax、Caspase-3蛋白的表达来发挥抗凋亡作用,从而对大鼠RIRI起到保护作用。

Abstract:Objective To investigate the effect of sangntodoxa cuneata extracts (SCE) on the expression of apoptosis related factors Bcl-2, Bax and Caspase-3 in rats with retinal ischemia-reperfusion injury (RIRI).Methods Ninety healthy and clean adult SD rats were randomly divided into control group, RIRI group and sargentodoxa cuneata extracts (SCE) group, 30 rats in each group. Then the 6 rats were sacrificed at 6h, 12h, 24h, 48h and 72h during reperfusion time for experiment. Nissl staining was used to observe the morphological changes of neural cells in each layer of retina in each group, and immunohistochemistry was used to detect the expression of apoptosis related factors Bcl-2, Bax and Caspase-3 in each layer of retina before and after SCE treatment.Results Nissl bodies were abundant, stained deeply and distributed in blocks in the control group. In the RIRI group, the nissl bodies were less than those in the control group, with light staining and uneven distribution. However, the staining of SCE group was better than that of the RIRI group. In the control group, the expression of Bcl-2 protein was almost non-existent, and the positive expression began to increase at 24 h after reperfusion, reaching the peak at 48 h after reperfusion. The positive expression of the SCE group was higher than that of the RIRI group at according time point (all P<0.05). Bax protein was slightly expressed in the control group, and the positive expression increased continuously after 12 h of reperfusion, reaching the peak at 24 h. The positive expression of SCE group was lower than that of the RIRI group at according time point (all P<0.05). In the control group, the expression of Caspase-3 protein was almost non-existent, it began to increase at 6h after reperfusion, reaching the peak at 12h. The positive expression in the SCE treatment group was lower than that in the model group in every period (all P<0.05).Conclusion SCE may up-regulate the expression of Bcl-2 protein and down-regulate the expression of Bax and Caspase-3 protein, so as to play the role of anti-apoptosis and protect the retinal ischemia-reperfusion injury in rats.
