摘    要:目的:建立用高效液相色谱法(HPLC)法测定山核桃树叶中球松素查耳酮(DHMC)含量的方法。方法:采用Kromasil C18(250 mm×4.6 mm,5μm)色谱柱,以甲醇-水(65∶35,V/V)为流动相,柱温为30℃,流速为1.0 ml/min,检测波长为342 nm,对DHMC的含量进行测定。结果:DHMC进样量在0.02~0.10μg范围内线性关系良好(r=0.999 6,n=6),精密度RSD为1.1%(n=6),重复性试验RSD为0.9%(n=6),加样回收率为101.6%(RSD=1.8%,n=6),经测定山核桃叶干品中DHMC的含量较高,达0.79%。结论:所建立的HPLC法专属性强,精密度好,结果准确。

Abstract:Objective: To establish the HPLC method for the determination on the content of DHMC in the Carya cathayensis Sarg..Methods: The Kromasil C18(250 mm×4.6 mm,5 μm) column was used for the determination on the content of DHMC with mobile phase consisted of methanol and water(65:35,v/v).Column temperature was at 30℃,flow velocity was 1.0 ml/min,the detective wavelength was at 342 nm.Results: The standard curves of DHMC was linear in the concentration range of 0.02-0.10 μg(r=0.999 6,n=6).The precision RSD was 1.1%(n=6),the repeatability RSD was 0.9%(n=6).The average recovery was 101.6%(RSD=1.8%,n=6),the content of DHMC in Carya cathayensis Sarg.was 0.79%.Conclusion: This method is simple and accurate with good reproducibility.
