实验动物瘤周注射转TNF-α基因的舌癌DNL在体内分布的研究

目的　探讨肿瘤周围注射经TNF α基因转导的舌癌引流淋巴结淋巴细胞 (DNL)体内分布的特点。方法　在建立人舌癌原位移植模型基础上 ,用氚标胸腺嘧啶核苷 (3H TdR)标记转TNF基因的舌癌DNL后肿瘤周围注射 ,分别检测注射后 6h、1 2h、2 4h、48h、1 68h肿瘤组织及各脏器单位重量的放射活性 (衰变率 ,DPM)。结果　各重要脏器及血液中单位重量的TNF/DNL在不同的时间由高到低依次是 :1 2h瘤、脾、肝、肺 ,2 4、48、1 68h瘤、脾、肺、肝。瘤体单位重量DPM最高 ,平均是正常组织的 1 0 .9倍 ,是外周血的 1 2 .5倍。结论　肿瘤部位注射可提高肿瘤局部的TNF/DNL细胞量 ,可望发挥更好的抑瘤效果

IN VIVO DISTRIBUTIONS OF DNL FROM TONGUE CANCER TRANSDUCED WITH TNF-α GENE AFTER LOCAL INJECTION IN TUMOR-BEARING MICE

Objective TO investigate the distribution of TNF gene transduced DNL from human tongue cancer after local injection in nude mice bearing human tongue carcinoma by orthotopic transplantation. Methods 3HTdR labeled DNL modified with TNF α gene were transferred in the murine tumor models by local injection and the relative radioactivity (DPM / g) in tumor and normal tissues was detected after 6h, 12h, 24h, 48h and 168h. Results Of the different organs and blood sample, 3H activity per gram at 12h was greatest in spleen, followed by liver and lung. 24h after infusion, 3H labeled DNL was still greatest in spleen, followed by lung and liver. In addition, the number of 3H labeled DNL was average higher than that of any other sample, 3H activity per gram in tumor tissue was 10.9times greater than that of normal tissue and 12.5times greater than that of blood sample. Conclusion The results indicate localization of DNL from human tongue cancer into tumor site was possibly increased by locoregional injection, and local injection of DNL modified with TNF gene can exert a better antitumor effect.
