Bcl-3在人乳腺癌组织中的表达及对乳腺癌MDA-MB-231细胞增殖与凋亡的影响

摘    要:目的: 探讨Bcl-3在乳腺癌中的表达及沉默后对乳腺癌细胞生物学行为的影响。方法: 选取80例乳腺癌组织及相应癌旁组织,采用免疫组织化学法检测其中Bcl 3蛋白表达,并分析癌组织中Bcl-3表达与临床病理参数间的关系;将乳腺癌MDA-MB-231细胞随机分为空白对照组(常规培养),阴性对照组(转染对照siRNA)和Bcl 3 siRNA组(转染Bcl-3 siRNA)。转染48 h后,免疫印迹法检测各组Bcl-3表达;MTT、平板克隆实验及流式细胞术分别检测各组细胞增殖能力、克隆形成能力和细胞凋亡。结果: Bcl-3蛋白在乳腺癌及相应癌旁组织中阳性表达率分别为72.5%和51.3%。乳腺癌组织中Bcl-3蛋白表达与组织学分级、淋巴结转移及Her 2表达呈正相关(P<0.05),而与年龄、肿瘤大小、病理分型及雌、孕激素受体无关(P>0.05)。空白对照组和阴性对照组Bcl-3表达水平明显高于Bcl 3 siRNA组(P均<0.01)。与空白对照组和阴性对照组比较,Bcl-3 siRNA组细胞增殖率和克隆形成率降低,细胞凋亡率增高(P均<0.01)。结论: Bcl 3在乳腺癌组织中呈高表达,其沉默后起抑癌作用,乳腺癌细胞增殖能力及克隆形成率降低、凋亡率增高。

Abstract:Objective To explore Bcl-3 expression in human breast cancer and its effect on breast cell proliferation and apoptosis. Methods Bcl 3 expression in 80 breast cancer tissues with corresponding adjacent tissues were detected by immunohistochemical staining. MDA MB 231 cells were randomized into blank control group(routine culture), negative control group(transfected with control siRNA) and Bcl-3 siRNA group(transfected with Bcl 3 siRNA); Bcl 3 expression was detected by western blotting. MTT assay, plate clone formation assay and flow cytometry were used to determine the cell proliferation, cloning formation efficiency and apoptosis rate, respectively. Results The positive rate of Bcl 3 expression in breast cancer tissue was 72.5%, while in adjacent tissues was 51.3%. The Bcl-3 expression in breast cancer tissues was positively correlated with histological grade, lymph node metastasis and Her 2 expression(P<0.05). Bcl-3 expression in blank control group and negative control group was significantly higher than in Bcl 3 siRNA group(P<0.01). Compared with blank control group and negative control group, Bcl 3 siRNA group showed lower cell proliferation rate, lower colony forming efficiency, higher apoptosis rate (P<0.01). Conclusion Bcl-3 expression in breast cancer tissues was high, and silencing it could suppress the growth of MDA MB 231 breast cancer cells by reducing cell proliferation, inhibiting cloning formation, and inducing cell apoptosis.
