人类白细胞抗原-G基因外显子8中14bp多态性与信使核糖核酸表达关系研究

摘    要:目的:分析正常妊娠妇女人类白细胞抗原-G(human leucocyte antingen-G,HLA-G)基因外显子8中14bp插入或缺失多态性分布频率,探讨14bp多态性与HLA-G异构体信使核糖核酸(messenger RNA,mRNA)表达的关系。方法:从144例正常妊娠妇女蜕膜组织中提取基因组DNA及总RNA。PCR扩增产物经SDS-PAGE后,分析外显子8中14bp多态性的分布频率;RT-PCR分析14bp / 、14bp-/-基因型与HLA-G异构体mRNA表达的关系。结果:144例正常妊娠妇女中14bp基因型分布频率,分别为14bp-/-(占12.5%),14bp /-(占56.3%),14bp / (占31.2%)。其中基因型为14bp / ,14bp-/-多态性影响HLA-G异构体mRNA的表达。结果显示,基因型14bp / 能稳定HLA-G3异构体mRNA的表达,但对HLA-G1无影响。结论:HLA-G基因外显子8中14bp多态性能影响HLA-G mRNA的表达。

Abstract:Objective:To investigate distribution frequency of the 14bp insertion/deletion polymorphism in HLA-G exon 8 and it's relationship between 14bp genotype and HLA-G isoform mRNA expression.Methods:Genomic DNA was extracted from 144 normal pregnancy decidual tissues. PCR was performed and then the PCR products and the 14bp insertion/deletion genotype polymorphism was analyzed by SDS-PAGE. Total RNA of the corresponding samples with 14bp / ,14bp-/-genotype was extracted and performed with RT-PCR to study the association of 14bp / , 14bp-/-genotype with HLA-G isoform mRNA expression.Results: Genotype distribution of the 14bp insertion/deletion in the 144 normal pregnancy decidual tissues is 14bp-/-(12.5%),14bp /-(56.3%),14bp / (31.2%) respectively, and genotype 14bp-/-,14bp / effects HLA-G3 isofrom expression, while no change was observed to the HLA-G1 isoform mRNA expression.Conclusion: Polymorphism of 14bp insertion/deletion in HLA-G exon 8 involves HLA-G mRNA expression.
