清眩降压汤对自发性高血压模型大鼠肾脏的保护作用

目的研究清眩降压汤对自发性高血压(SHR)大鼠血压的影响和肾脏保护作用。方法 40只SHR大鼠随机分为清眩降压汤高剂量组(5.372g/d)、低剂量组(1.7g/d),替米沙坦组(2.4mg/d)和模型组(予矿泉水),每组10只;另设10只Wistar大鼠为正常组(正常饲养);连续给药8周。给药前,单次给药2h、6h、12h、24h,给药3天、1周、4周、8周,分别检测各组大鼠收缩压和舒张压;最后一次检测血压后10%水合氯醛麻醉取血,酶联免疫法和放射免疫法检测大鼠血清中血管紧张素Ⅱ(AngⅡ)和血管紧张素1-7(Ang1-7)的含量;HE染色观察大鼠肾脏组织的病理改变;免疫组织化学方法检测AngⅡ在肾脏的表达。结果清眩降压汤给药1次,各时间点大鼠血压未出现明显变化,替米沙坦组大鼠给药后2h收缩压、舒张压均明显下降(P<0.05);连续治疗8周后,未发现清眩降压汤高、低剂量组大鼠血压明显变化,替米沙坦组大鼠收缩压显著下降(P<0.05),而模型组大鼠收缩压升高显著(P<0.05或P<0.01),AngⅡ和Ang1-7在血压升高大鼠中较血压正常大鼠水平要低(P<0.05),各给药组大鼠肾脏肾小球和肾小管增生程度较模型组大鼠均得到明显抑制。结论清眩降压汤可以控制SHR收缩压的进一步升高,抑制由于血压升高导致的大鼠肾小球和肾小管增生,对SHR有潜在的肾脏保护作用。

Protective Effect of Qingxuan Jiangya Decoction on Kidney of Spontaneous Hypertension Rats

Objective To study the protective effect of Qingxuan Jiangya Decoction(Decoction for treating dizziness and lowering blood pressure) on blood pressure and kidney of spontaneous hypertension rats(SHR).Methods Forty SHR were randomized into 4 groups,i.e.Qingxuan Jiangya Decoction high-dose group and low-dose group,accepted Qingxuan Jiangya Decoction 5.372g/d and 1.7g/d respectively,the telmisartan group,accepted telmisartan 2.4mg/d,and the model group,with 10 rats in each.Ten Wistar rats w ere taken into normal control group.The rats were treated with intragastric administration for 8 weeks in succession.Blood pressure was determined respectively before treatment,2h,6h,12h,24h after first treatment and 3d,1 week,4 weeks and 8 weeks treatment.After the last blood pressure examination,10% chloral hydrate anesthesia was adopted to collect blood for enzyme linked i mmunosorbent assay(ELISA) and radioimmunoassay(RIA) tests to measure the Ang Ⅱ and Ang1-7 contents.HE staining was used to observe the pathological chan ges of kidney tissue,and the immunohistochemical method was applied to determine the AngⅡ expre ssion in the kidney.Results With the first time intragastric administration,blood pressure in the high-dose and low-dose of Qingxuan Jiangya Decoction was not signi ficantly changed,while the systolic and diastolic pressure of the telmisartan group was signi ficantly lowered(P<0.05) 2 hours after treatment.After the treatment for 8 weeks in succession,blood pressure in the high-dose and low-dose Qingxuan Jiangya Decoction groups was not significantly changed(P>0.05),the systolic pressure of the telmisartan group was significantly lowered(P<0.05),and the systolic pressure of the model group was increased significantly(P<0.05 or P<0.01).Ang Ⅱ and Ang1-7 contents in the SHRs were low er than in the normal rats(P<0.05);and the glomerular and tubular hyperplasia in all treatment groups were both significantly inhibited compared with those of the model group.C onclusion Qingxuan Jiangya Decoction is effective in controlling the further increase of SHR s ystolic pressure,inhibitying the hypertensive glomerular and tubular hyperplasia,thus providing a potential kidney protection for SHR.
