摘    要:目的:分析miR-129-5p和信号转导与转录激活因子3(STAT3)在子宫内膜癌(EC)中的表达及其临床意义。方法:选择2014年6月至2015年5月我院收治的子宫内膜癌患者54例,手术切除癌组织及癌组织边缘≥5 cm处的癌旁组织分别为子宫内膜癌组、对照组,收集患者的临床病理资料,qRT-PCR检测miR-129-5p与STAT3 mRNA表达水平,免疫组化法检测STAT3蛋白表达,采用Pearson法进行相关性分析,Kaplan-Meier法分析子宫内膜癌患者5年的生存情况。结果:qRT-PCR检测结果显示子宫内膜癌组miR-129-5p表达水平异常低于对照组,而STAT3表达水平高于对照组,差异均具有统计学意义(P<0.05);免疫组化显示STAT3定位于细胞核和细胞浆,子宫内膜癌组STAT3蛋白阳性率显著高于对照组(P<0.05);观察miR-129-5p、STAT3与子宫内膜癌患者临床病理特征的关系,结果显示miR-129-5p与病灶直径、分化程度、浸润深度相关,STAT3与TNM分期、分化程度及浸润深度相关;Pearson相关性分析显示miR-129-5p与STAT3呈显著负相关性;Kaplan-Meier分析结果显示miR-129-5p、STAT3表达与子宫内膜癌患者PFS、OS明显相关,miR-129-5p高表达组PFS(38.26%)、OS(51.33%)高于低表达组PFS(15.27%)、OS(21.23%),STAT3阴性表达组PFS(40.26%)、OS(52.19%)高于阳性表达组PFS(20.51%)、OS(37.31%),差异均具有统计学意义(P<0.05)。结论:miR-129-5p在子宫内膜癌组织中异常低表达,STAT3在子宫内膜癌组织中显著高表达,miR-129-5p低表达增加患者预后不良风险,STAT3高表达增加患者预后不良风险。

Abstract:Objective:To analyze the expressions and clinical significances of miR-129-5p and signal transduction and activators of transduction 3 (STAT3) in endometrial cancer (EC) tissues.Methods:54 patients with endometrial cancer admitted to our hospital from June 2014 to May 2015 were selected.Cancer tissues and adjacent tissues ≥5 cm from the margin of cancer tissues surgically removed were set as endometrial cancer group and control group respectively.The expressions of miR-129-5p and STAT3 were detected by qRT-PCR.The expression of STAT3 protein was detected by immunohistochemistry.Pearson method was used for correlation analysis.Kaplan-Meier method was used to analyze the 5-year survival of patients with endometrial cancer.Results:The results of qRT-PCR showed that the expression of miR-129-5p in endometrial cancer group was lower than that in control group.And the expression level of STAT3 was higher than that of the control group.And the differences were statistically significant (P<0.05).Immunohistochemistry showed that STAT3 was located in the nucleus and cytoplasm,and the positive rate of STAT3 protein in endometrial cancer group was significantly higher than that in control group (P<0.05).MiR-129-5p was correlated with lesion diameter,differentiation degree,and depth of invasion.STAT3 was correlated with TNM stage,differentiation degree,and infiltration depth.Pearson correlation analysis showed that there was a significant negative correlation between miR-129-5p with STAT3.Kaplan-Meier analysis showed that the expressions of miR-129-5p and STAT3 were significantly correlated with PFS and OS in patients with endometrial cancer.PFS (38.26%) and OS (51.33%) in the miR-129-5p high expression group were higher than PFS (15.27%) and OS (21.23%) in the low expression group.PFS (40.26%) and OS (52.19%) in STAT3 negative expression group were higher than PFS (20.51%) and OS (37.31%) in positive expression group,and the differences were statistically significant (P<0.05).Conclusion:The expression of miR-129-5p is abnormally low in endometrial cancer.STAT3 is significantly highly expressed in endometrial cancer.Low expression of miR-129-5p increases the risk of poor prognosis,while high expression of STAT3 increases the risk of poor prognosis.
