葛根异黄酮酶解的研究

摘    要:为了提高葛根异黄酮的生物利用率,用黑曲酶Aspergillusnigersp. 0848菌株生成的酶水解葛根异黄酮,去除了葛根异黄酮上的部分糖基使其生成相应的活性较高的甙元。通过HPLC分析,以酶解前后葛根中大豆甙元和染料木素的峰面积之比为指标,对酶解条件进行了考察。结果表明,葛根在37℃,pH=5 0,反应时间24h条件下,酶解最为彻底。同时考察了酶解产物的HPLC分离条件,结果表明,在流动相中加入适量醋酸可以使酶解产物得到有效分离。

Abstract:The enzyme produced by Aspergillus niger sp.0848 strain was used to hydrolyze the isoflavones of kudzu root in order to improve the bioavailability.The sugar moieties were removed and more active free aglycones were produced.By comparison of the peak area ratios of daidzein and genistein after/before the enzyme hydrolysis in HPLC analysis,the hydrolysis conditions were examined.The optimal conditions were pH 5.0 at 37 ℃ for 24 h.The products of enzyme hydrolysis were efficiently resolved when acetic acid was added to the mobile phase in HPLC analysis.
