摘    要:利用原子力显微镜(Atomic Force Microscopy,AFM)对淋巴细胞表面形貌进行了形态学的初步研究,观察到了其膜表面其他显微技术所不能发现的超微结构.同时也运用扫描近场光学显微镜(Scanning Near field Optical Microscopy,SNOM)对淋巴细胞进行成像,观察了其对光的透射、吸收等光学性质,并对两种成像方法进行了比较.研究发现:淋巴细胞膜表面凹凸不平,分布着大量直径几十到几百纳米不等的小颗粒;淋巴细胞中央部位有自发荧光现象.结果表明,AFM和SNOM可作为进一步探讨淋巴细胞的结构与功能关系的有力工具.

Abstract:The atomic force microscope (AFM),with its unique high-resolution capability, plays an ever-increasingly important role in the field of physical,chemical and biological research.The present study applied AFM to the observation of the single lymphocyte cell,its part and the ultratructure of its membrane surface.The result of this study suggests that the newly established method of specimen preparation may provide high quality three-dimensional images of the cells as well as their surface fine structures.Some structures that are difficult to be found by other microscopy also can be observed with AFM.At the same time,we also used the scanning near field optical microscopy (SNOM) to study these cells with their optical property such as transmission and absorbency.The result shows that there are many particles on the lymphocyte membrane surface,the diameter of them are about a hundred nano and the autofluorescence of lymphocyte was observed by SNOM.Comparing resolution of the both techniques,we found that SNOM can offer more optical information about the cells than other microscopy.And this information maybe helps us to realize the relationship of the ultratructure and the biochemical function.As a conclusion,both AFM and SNOM can be used as useful techniques to study the lymphocyte cells.
