温阳消饮法对胸腔积液豚鼠胸膜间皮细胞水通道蛋白1表达的影响

摘    要:目的:研究温阳消饮法消除胸液与调节水通道蛋白表达的相关性。方法:将健康非纯种短毛成年豚鼠随机分为A-温阳消饮组、B-模型+生理盐水组、C-模型组、D-空白组,各实验组再根据造模后第1、2、3、5、7、10、14 d随机分为7个小组。1%角叉菜胶胸腔注射建立胸腔积液模型。造模后2h给药,连续14 d。分别于造模后第1、2、3、5、7、10、14 d处死各组豚鼠。计量胸液,并用免疫组化法检测脏层与壁层胸膜间皮细胞水通道蛋白1(AQP1)表达。结果:①A5组较B5、C5组,A7组较B7、C7组胸液量减少(P0.05);②A7组脏层胸膜间皮细胞AQP1表达较B7、C7组增强(P0.05);③A2、A3、A5、A7、A10、A14组分别与B2、B3、B5、B7、B10、B14组及C2、C3、C5、C7、C10、C14组比较,其壁层胸膜间皮细胞AQP1表达减弱,差异有统计学意义(P0.05);④A2、A3、A5、A7、A10、A14各组脏层胸膜间皮细胞AQP1表达强于壁层胸膜(P0.05),B7、B10、B14组及C7、C10、C14组脏层胸膜间皮细胞AQP1表达弱于壁层胸膜(P0.05)。结论:温阳消饮法能减少胸腔积液模型豚鼠胸液量,其作用机理可能是:①上调脏层胸膜间皮细胞AQP1表达,有助于胸液转运;②下调壁层胸膜间皮细胞AQP1表达,减少胸液形成。

Influence on Variation of Aquaporin 1 Expression in Pleural Mesothelial Cells of Pleural Effusion Guinea Pig Treated with Warm Yang to Resolve Fluid-retention Therapy

Abstract:Objective: To explore the correlation between pleural effusion removal by warming Yang to resolve fluid-retention therapy and expression of aquaporin regulation.Methods: Non-purebred short-haired healthy adult guinea pigs were randomized into four groups including group A(the treatment group with warming yang to resolve fluid-retention therapy),group B(the model group treated by physiological saline),group C(the model group) and group D(control group),which were further divided randomly into seven subgroups in accordance with the days of 1,2,3,5,7,10 after molding.The pleural effusion mold was formulated by 1% Carrageenan injection.The treatment was applied in two hours after molding for incessant 14 days.Then the rats were killed at 1,2,3,5,7,10 and 14 days after molding.The pleural effusion was firstly measured,while the AQP1 expression in parietal or visceral pleural mesothelial cells was tested with immunohistochemistrical method.Results:①Compared with group B5 and C5,pleural effusion in rats of group A5 was significantly reduced(P<0.05);Compared with group B7 and C7,pleural effusion of group A7 was notably reduced(P<0.05).②Compared with group B7 and C7,AQP1 expression of visceral pleural mesothelial cells in group A7 was significantly enhanced(P<0.05).③With the exception A1,thr AQP1 expression in parietal pleural mesothelial cells was statistically lowered and different from that in B2,B3,B5,B7,B10,B14 or C2,C3,C5,C7,C10,C14(P<0.05).④The AQP1 expression of visceral pleural mesothelial cells in A2,A3,A5,A7,A10 and A14 was stronger than that in parietal ones(P<0.05);and that in B7,B10,B14 and C7,C10,C 14 was weaker than that in parietal ones(P<0.05).Conclusion: Warming Yang to resolve fluid-retention therapy reduced the volume of the pleural effusion in molding rats,as its underlying mechanism might be:①raising the AQP1 expression in visceral pleural mesothelial cells facilitates the transportation of pleural effusion;②weakening the AQP1 expression in parietal pleural mesothelial cells help form less pleural effusion.
