金线莲菌根真菌的分离和鉴定

摘    要:目的分离及鉴定野生金线莲根内菌根真菌。方法采用平皿点种法进行金线莲菌根真菌的分离,提取菌株的总DNA,采用丝状真菌ITS通用引物对其rDNA相应区段进行PCR扩增,产物经纯化后测序。结果结合各菌株的形态特征和ITS区域基因序列特征,将A12菌株、5号菌株、2号菌株、6号菌株、A46菌株和A55菌株鉴定为丝核菌(Rhizoctonia sp.),其有性态为Thanatephorus sp.或Ceratobasidium sp.。结论通过对金线莲菌根真菌的分离鉴定,有望将其应用到金线莲及兰科其它濒危药用植物繁殖中去。

Isolation and Identification of mycorrhizal fungus from Anoectochilus roxburghii

Abstract:Objective Isolation and identification the mycorrhizal fungi from natural Anoectochilus roxburghii ( Wall. ) Lindl. Method To isolate the myeorrhizal fungi from natural A. roxburghii ( Wall. ) Lindl. by plate cultivation. Total DNA of fungi were extracted and amplified by using universal primers ITS1 and ITS4, products were purified and sequenced. Result Based on the morphological characteristics of the various strains and the ITS region gene sequence, strain A12, strain 5, strain 2, strain 6, strain A46 and strain A55 were identified as Rhizoctonia sp. and their sexual state is Thanatephorus sp. or Ceratobasi- dium sp. Conclusion Through the isolation and identification the myeorrhizal fungi from natural A. rox- burghii ( Wall. ) Lindl. , it is possible to apply them in the cultivation of medicinal plants (Orehidaceae) and other endangered medicinal plants.
