| 摘 要： | 目的探讨靶向血管内皮细胞生长因子受体3(vascular endothelial growth factor receptor-3,VEGFR-3)基因的小干扰RNA(small interfering RNA,siRNA)腺病毒载体对人结肠癌Lo Vo细胞系凋亡及侵袭的影响。方法将靶向VEGFR-3 siRNA腺病毒转染结肠癌Lo Vo细胞,以Western blotting检测VEGFR-3蛋白的表达,Hoechst 33342染色法和流式细胞仪检测Lo Vo细胞的凋亡情况,用Transwell小室测定Lo Vo细胞的侵袭力。结果实验组与空白对照组和阴性对照组比较,实验组中转染靶向VEGFR-3 siRNA腺病毒后结肠癌Lo Vo细胞中VEGFR-3蛋白的表达被下调(P0.05)。Hoechst 33342染色法和流式细胞仪检测Lo Vo细胞凋亡率明显升高(P0.05),Transwell小室测定Lo Vo细胞侵袭能力下降(P0.05)。结论靶向VEGFR-3 siRNA腺病毒能下调结肠癌LoVo细胞中VEGFR-3蛋白的表达,从而诱导结肠癌Lo Vo细胞的凋亡,抑制其侵袭能力,因此,VEGFR-3可以作为结肠癌靶向治疗的一个潜在靶点。 |
