蔷薇红景天的化学成分及对紫外线诱导的A375-S2细胞死亡的抑制作用

摘    要:目的对蔷薇红景天(Rhodiola roseaL.)干燥根茎的体积分数为30%乙醇提取物化学成分进行研究,并探讨其中部分单体化合物对紫外线诱导的A375-S2细胞死亡的抑制作用。方法利用硅胶柱色谱等多种方法进行分离纯化,根据理化性质和NMR、MS等波谱数据进行结构鉴定;采用MTT法检测细胞的存活率来评价化合物对紫外线照射诱导的细胞死亡的保护作用。结果分离得到7个化合物,经鉴定分别为:迷迭香酸(rosmarinci acid,1)、山柰酚(kaempferol,2)、山柰酚-7-O-鼠李糖苷(kaempferol 7-O-rhamnopyranoside,3)、草质素-7-O-α-L-鼠李糖苷(herbacetin 7-O-α-L-rhamnopyranoside,4)、草质素-7-O-(3″-O-β-D-葡萄糖)-α-L-鼠李糖苷(herbacetin 7-O-6-(3″-O-β-D-glucopyranosyl)-α-L-rhamnopyranoside,5)、槲皮素(quercetin,6)和芦丁(rutin,7)。结论化合物1为首次从红景天属植物中分离得到,化合物7为首次从该种植物中分离得到;化合物5对紫外线诱导的A375-S2细胞死亡具有明显的抑制作用,且对该细胞没有显示出细胞毒活性。

Abstract:Objective To elucidate the structures of the constituents from the 30% ethanol extract of Rhodiola rosea L.and invest the effects of the constituents on UV-induced A375-S2 cell death.Methods Column chromatography methods and so on were used for isolation of chemical constituents,physico-chemical characters and spectroscopic evidences were employment for structural identification.The inhibitory effect of the part flavonoids from the extract on UV-induced A375-S2 cell death was estimated by the viability of the cell,the viability was measured by MTT assay.Results Seven compounds were isolated,and identified as rosmarinci acid(1),kaempferol(2),kaempferol 7-O-rhamnopyranoside(3),herbacetin 7-O-α-L-rhamnopyranoside(4),herbacetin 7-O-(3″-Oβ-D-glucopyranosyl)-α-L-rhamnopyranoside(5),quercetin(6) and rutin(7),respectively.Conclusions Compound 1 is isolated from Rhodiola genus for the first time;Compound 7 is isolated from Rhodiola rosea L.for the first time;Compound 5 shows the inhibitory effect on UV-induced A375-S2 cell death and no cytotoxic effect on the normal A375-S2 cells.
