| 摘 要： | 重组表达菌株的稳定性是决定试剂盒抗原生产的关键因素,对本室构建的重组菌株rNS- 3ABplysS携带质粒和表达蛋白的稳定性进行了研究,在含Amp的斜面连续传1～10代时,分别作了检测连续传代质粒、携带质粒DNA、抗原蛋白表达的稳定性试验。发现各菌株携带质粒稳定性较好,酶切及电泳图谱未有明显改变,且外源目标蛋白可稳定表达,可作为理想菌株。

| Abstract: | The stability of the recombinant expressing bacterium is a key factor of the antigen kit.The sta- bility of the recombinant bacterium rNS-3ABplysS carried plasmid and expression protein was studied.The re- suit showed that the plasmid carried by the bacterium after continuous passage on Amp~ slope was stable,differ- ent generation carrier plasmids digested by endonuclease were nearly the same as the original plasmid digested, and extrinsic target protein could be stably expressed.So it was an ideal bacterium. |
