银杏叶提取物对肺纤维化模型大鼠肺组织MMP-2mRNA表达的影响

摘    要:目的观察银杏叶提取物对肺纤维化大鼠不同时间点肺组织MMP-mRNA表达的影响。方法96只健康Wist~大鼠随机分为正常对照组(A组)、模型组(B组)、强的松组(c组)和银杏叶提取物干预组(D组),每组各24只,除A组外,其余各组采用博莱霉素复制大鼠肺纤维化模型。造模第2天起,D组用银杏叶提取物灌胃,C组用醋酸强的松混悬液灌胃,A、B组用生理盐水灌胃。每组分别于造模后第7、14、28天随机取8只大鼠,处死后采集肺组织,抽提RNA,采用RT—PCR法得到MMP-2mRNA产物,用琼脂糖凝胶电泳和图像分析方法观察MMP4mRNA的表达。结果各实验组在各时间点均有MMP4mRNA的表达,与A组相比,B组第7天含量显著升高(P〈0.01),第14天时达到高峰,第28天虽较第7、14天时有所降低,但仍明显高于同期A组(P〈0.01)。与B组比较,C、D组各时间点含量均明显降低(P〈0.01)。结论造模后各组MMP-2mRNA表达增强,银杏叶提取物能够有效抑制MMP-mRNA的过度表达,从而对肺损伤和肺纤维化起到一定的防治作用。

Abstract:Objective To observe the effects of ginkgo biloba extract (GBE) on expression of MMP-2mRNA at different time points in rats with experimental pulmonary fibrosis. Methods Ninety-six healthy Wistar rats were randomly divided into a normal control group (Group A), a model group (Group B), a prednisone group (Group C) and a GBE group (Group D), 24 cases in each one. The bleomycin was applied to establish the rat model of pulmonary fibrosis in all groups except the Group A. From two days after the model establishment, intragastric administration of GBE was used in Group D, that of prednisone acetate was in Group C and that of normal saline was in Group A and Group B. 7, 14, 28 days after the model establishment, 8 cases of rats were randomly selected in each group to sacrifice and lung tissue was acquired, in which RNA was abstracted. The RT-PCR method was adopted to obtain the products of MMP-2mRNA, while agarose gel electrophoresis and image analysis method was adopted to observe the expression of MMP- 2mRNA. Results The expression of MMP-2mRNA was positive in each experiment group at all different time points. Compared with Group A, the level of MMP-2mRNA in Group B was significantly increased on the 7th day (P 〈0. 01 ) and reached the highest on the 14~ day; although on the 28th day the level was lower compared with that on the 7~ and 14'h day, it was obviously higher than that in the Group A (P 〈0. 01 ). Compared with Group B, the level in Group C and Group D at each time point was significantly decreased (P 〈 0. 01 ). Conclusion After the model establishment the expression of MMP-2mRNA was enhanced in each group, indicating that GBE could effectively prohibit the over-expression of MMP-2mRNA, which could play a prevention and treatment role for lung injury and pulmonary fibrosis.
