地塞米松对缺氧缺血性脑损伤新生大鼠脑组织AQP-4表达的研究

摘    要:目的探讨DEX(地塞米松)对缺氧缺血性脑损伤新生鼠脑组织AQP-4表达的影响,为临床治疗新生儿缺氧缺血脑病提供实验和理论依据,揭示其发病机制,以期指导该病的早期治疗。方法将健康7日龄SD新生大鼠96只,清洁级,雌雄不限,平均体重12-18g,随机分为假手术组,HIBD组,DEX组,各组32只,分离左侧颈总动脉并结扎,吸入92%N2+8%O2氮氧混合气体行缺氧处理3h,制成HIBD模型。假手术组只作分离左侧颈总动脉而不结扎,不作缺氧处理,不给与药物处理。DEX治疗组于建模成功后立即腹腔注射10mg/kg,假手术组及HIBD组大鼠腹腔注射等量生理盐水,上述各组均于术后6h、24h、48h、72h随机抽取8只,于麻醉下处死取脑组织,通过Western blot方法检测新生大鼠脑组织AQP-4蛋白表达及实时定量PCR检测AQP-4 mRNA的表达。结果 SD鼠在缺氧处理后出现异常行为学改变。AQP-4 mRNA及蛋白的表达在缺氧诱导后显著升高,DEX的治疗则使SD鼠异常行为的表现程度减轻,后趋于正常,且能够降低HIBD导致的AQP-4的升高。结论 DEX对HIBD的治疗具有一定的疗效。

Abstract:Objective: It is to discuss whether DEX (dexamethasone) have a certain curative effect on hypoxic ischemic brain damage in newborn rat, in order to guide early treatment of hypoxic-ischemic encephaiopathy. Methods: 96 healthy neonatal SD rats of 7-days old were randomly divided into control group, HIBD group, DEXgroup, and 32 of them were assigned to each group. Each group was randomly divided into some corresponding time of postoperation 6h, 24h, 48h and 72h which was measured the mRNA and protein expression of AQP-4 at each time spot. Results: Abnormal behavioral of SD rats emerged after anaerobic treatment. AQP-4 mRNA and protein expression significantly increased after anoxic induction, while the treatment of DEX will redress abnormal behavioral of SD rats and reduce the rise trend of AQP-4 in HIBD group. Conlusion: The therapeutic effect of DEX treatment group was obviously better than HIBD group.
