冠心Ⅰ号治疗冠心病的临床和实验研究

摘    要:目的:研究冠心Ⅰ号的临床疗效和作用机制。方法:应用具有益气养阴、活血通阳功效的冠心Ⅰ号对20例冠心病患者进行临床治疗观察,并用冠心Ⅰ号对具有动脉粥样硬化动物模型的大鼠灌胃,观察其对主动脉形态及对主动脉MMP-9蛋白、MMP-9/TIMP-1蛋白比值表达的影响。结果:冠心Ⅰ号治疗心绞痛总有效率为90%;临床中医证候总疗效总有效率为95%;心电图总有效率为45%。动物实验表明:冠心Ⅰ号可改善动脉粥样硬化动物模型大鼠的血管内膜细胞增生,降低MMP-9的蛋白表达,调节MMP-9/TIMP-1失衡状态。结论:冠心Ⅰ号是通过降低MMP-9的蛋白表达,调节MMP-9/TIMP-1失衡状态,改善血管内膜细胞增生及心肌缺血,减轻动脉粥样硬化病变程度。

Abstract:Objectives:To study the clinical effects and therapeutic mechanisms of the application of Chinese traditional medicine,the Guanxin No.1 in the treatment of coronary heart disease.Methods:The clinical effects were observed in 20 patients with coronary heart disease,to whom Guanxin No.1 were given.The rats of atherosclerotic model were used as experimental models with cardiovascular disease.The rats were fed with Guanxin No.1 and the effects on the aorta shapes and expressions of MMP-9 protein and MMP-9/TIMP-1 of rats were noticed.Results:The total effective rate of angina,clinical characteristics of syndromes of TCM and ECG respectively was 90%,95%and 45% with Guanxin No.1.Experimental study showed that Guanxin No.1 could improve arterial smooth muscular cells cultured,reduce expressions of MMP-9 protein and adjust the state of MMP-9/TIMP-1 imbalance from the rats of atherosclerotic model.Conclusions:Guanxin No.1 could improve arterial smooth muscular cells cultured,myocardial ischemia and reduce the severity of coronary atherosclerosis by reducing expressions of MMP-9 protein and adjusting the state of MMP-9/TIMP-1 imbalance.
