摘 要: | 目的:建立桑叶药材中槲皮素、山奈酚、槲皮苷、芦丁、异槲皮苷、绿原酸、东莨菪内酯的含量测定方法。方法:采用HPLC外标法。采用色谱柱:AgilentC18柱(150mm×4.6mm,5μm),以乙腈为流动相A,以0.1%磷酸溶液为流动相B,梯度洗脱。检测波长分别切换为350nm,298nm,350nm,255nm,360nm,310nm。结果:槲皮素、山奈酚、槲皮苷、芦丁、异槲皮苷、绿原酸、东莨菪内酯的线性范围分别为0.0958~0.6065、0.0318~0.2016、0.1157~0.7326、0.1110~0.7032、0.1883~1.1923、0.2945~1.8652、0.1892~1.1979,平均回收率分别为102.8%、101.8%、99.5%、103.8%、99.1%、100.5%、98.7%。结论:本方法简便、准确,在同一色谱条件下可同时测定桑叶中槲皮素、山奈酚、槲皮苷、芦丁、异槲皮苷、绿原酸、东莨菪内酯的含量。

Abstract: | Objective:To determine the contents of quercetin、kaempferol、quercitrin、rutin、isoquercetin、chlorogenic acid and scopoletin in mulberry leaves by HPLC.Methods:Anexternal standard method was adopped by HPLC with Agilent C18 column(150mm ×4.6mm,5 μm)as fixed phase,The mobile phase A was acetonitrile and the mobile phase B was 0.1%phosphoric acid.The detection wavelength respectively was at 350nm、298nm、350nm、255nm、360nm、310nm.Results:Quercetin had a good linearity in the range of 0.0958~0.6065mg/ml.The average recovery rate was 102.8%.Kaempferol had a good linearity in the range of 0.0318~0.2016 mg/ml.The average recovery rate was 101.8%.Quercitrin had a good linearity in the range of 0.1157~0.7326 mg/ml.The average recovery rate was 99.5%.Rutin had a good linearity in the range of 0.1110~0.7032 mg/ml.The average recovery rate was 103.8%.Isoquercetin had a good linearity in the range of 0.1883~1.1923 mg/ml.The average recovery rate was 99.1%.Chlorogenic acid had a good linearity in the range of 0.2945~1.8652 mg/ml.The average recovery rate was 100.5%.Scopoletin had a good linearity in the range of 0.1892~1.1979 mg/ml.The average recovery rate was 98.7%.Conclusion:The method is simple and accurate.It can be used to determine quercetin 、kaempferol 、quercitrin、rutin、isoquercetin、chlorogenic acid and scopoletin together with the same chromatogram condition. |
