通心络对动脉硬化兔胸主动脉LOX-1表达干预的实验研究

目的:探讨通心络对动脉硬化兔胸主动脉植物血凝素样氧化低密度脂蛋白受体-1(LOX-1)表达的影响及机制。方法:将24只雄性新西兰大白兔随机分为正常饮食组、高脂饮食组及高脂饮食加通心络干预组,应用免疫组化测定兔胸主动脉中LOX-1的蛋白表达水平,用RT-PCR测定兔胸主动脉中LOX-1基因表达的水平。结果:高脂组及高脂饮食加通心络干预组胸主动脉中LOX-1蛋白及基因表达水平明显高于正常饮食组(P<0.05),通心络干预组胸主动脉中LOX-1蛋白及基因表达水平明显低于高脂饮食组(P<0.05)。结论:动脉硬化兔LOX-1蛋白及基因表达水平明显升高,通心络能够明显抑制动脉硬化兔LOX-1蛋白及基因表达。

The effect of tong Xin Luo on the expression of Lox-1 in aorta of atherosclerotic rabbits

Objective:To explore the effect of Tong Xin Luo on the gene and protein expression of lox-1 in aorta of atherosclerotic rabbits and in mechnism.methods:Twenty-four male New Zealand white rabbits were randomly divided into three groups:normal diet group、hypercholesterol diet group、hypercholesterol diet plus Tong Xin Luo group,the protein expression of lox-1 in aorta of rabbits was examined by immunohistochemistry,the gene expression of lox-1 in aorta of rabbits was examined by RT-PCR.Results:The gene and protein expression levels of lox-1 in hypercholesterol diet group and hypercholesterol diet plus Tong Xin Luo group were significant higher than those in of normal diet(P<0.05),the gene and protein expression levels of lox-1 in hypercholesterol diet plus Tong Xin Luo group were significant lower than those in hypercholesterol diet group(P<0.05).Conclusions:the gene and protein expression levels of lox-1 in aorta of atherosclerotic rabbits strongly increased,Tong Xin Luo could strongly inhibit the gene and protein expression in aorta of atherosclerotic rabbits.
