HER-2在宫颈病变脱落细胞的表达及临床意义

目的:研究癌基因HER-2在不同程度宫颈病变脱落细胞的表达,探讨它在宫颈癌发生发展中的作用及临床意义。方法:用荧光原位杂交(FISH)法检测12例正常宫颈、55例宫颈上皮内瘤变(CIN)及24例浸润性宫颈癌脱落细胞中HER-2的表达。结果:HER-2在正常宫颈脱落细胞中不表达,在CINⅠ、CINⅡ～Ⅲ及宫颈癌的阳性表达率分别为6.3%、25.6%及58.3%。HER-2在CINⅡ～Ⅲ及子宫颈癌组表达均显著高于正常组(P<0.01),同时宫颈癌组HER-2明显高于CIN组(P<0.01)。HER-2高表达与细胞分级明显相关(P<0.01),与临床分期、病理类型无关。结论:HER-2基因高表达在宫颈癌发生发展中可能起重要作用,在宫颈病变脱落细胞中检测HER-2基因对探讨宫颈病变的进展和预测不良预后可能有重要意义。

Expression and clinical significance of HER-2 oncogene in cervical lesion cells

Objective:To investigate the expression of the HER-2 oncogene in cervical lesion,and to evaluate the clinical significance of HER-2 expression in cervical lesion.Method:The expression of HER-2 in 24 cases with infiltrating cervical cancer,55 cases with cervical intraepithelial neoplasia(CIN) and 12 cases with normal control cervix were detected by the fluorescent in situ hybridization(FISH).Results:The expression of HER-2 was not observed in normal cervix cells.Positive expression rates of the HER-2 in CINⅠ,CINⅡ～Ⅲ and infiltrating cervical cancer cells were 6.3%,25.6% and 58.3% respectively.Compared with control group,HER-2 expression rate in CINⅡ～Ⅲ and infiltrating cervical cancer was different significantly(P<0.01),and there was a significantly difference between CIN group and infiltrating cervical cancer group(P<0.01).The over expression of HER-2 was correlated with the differentiation degree of carcinoma closely(P<0.01).No correlation was found between positive expression and clinical stages,nor pathologic types.Conclusion:The over-expression of HER-2 may play an important role in cervical carcinogenesis,it may be recommended to evaluate the progression and predict poor prognosis of cervical lesion.
