摘    要:目的探讨促红细胞生成素(EPO)对大鼠心肌缺血再灌注损伤(MIRI)的保护作用及其作用机制。方法采用结扎左冠状动脉前降支30min,再灌注120min的方法建立大鼠MIRI模型,将45只大鼠随机分为假手术组、MIRI组、EPO组(rHuEPO3000U/kg)。肢体Ⅱ导联心电图记录再灌注心律失常情况,检测血清丙二醛(MDA)、肌酸激酶(CK)、乳酸脱氢酶(LDH)水平。结果MIRI组和EPO组心律失常评分较假手术组明显升高(P〈0.05);EPO组室性心动过速和心室纤颤的持续时间明显低于MIRI组(P〈0.05)。MIRI组和EPO组血清MDA、CK、LDH水平较假手术组明显升高(P〈0.05);EPO组明显低于MIRI组(P〈0.05)。结论EPO对心肌MIRI有明显的保护作用,其机制可能与抗氧化应激有关。

Abstract:Objective To study the effects oferythropoietin(EPO)on myocardial ischemia-reperfusion inju-ry(MIPd)in rats.Methods The left anterior descending coronary artery was ligated for 30 minutes and then loosed for 120minutes to establish the rat model of myocardial MIRI.Forty five rats were randomly divided into sham-opera-tion group,MIRI group and EPO group(rHuEPO 3000U/kg).ECG was traced continuously to record the arrhythmia caused by reperfusion.The levds of serum MDA,CK and LDH were detected at reperfusion for 120min.Results The arrhythmia score in MIRI group and EPO group were significantly hisher than those in sham-operation group(P<0.05).Ventrlcular mchycardia and the duration of ventricular fibrillation in EPO group were significantly lower than those in MIRI group(P<0.05).The levels of serum MDA,CK and LDH in MIRI group and EPO group were signifi-cantly higher than those in sham-operation group(P<0.05).EPO group were significantly lower than MIRI group(P<0.05).Conclussion EPO had protective effects on myocardial MIRI and the mechanism might be related to anti-oxidative stress.
