儿童外周血不同类型白细胞柯萨奇-腺病毒受体的表达

摘    要:目的研究柯萨奇-腺病毒受体(CAR)在儿童外周血不同类型白细胞中的表达。方法采用全血免疫荧光流式细胞术检测南华大学附属第一医院儿科2005年1-6月不同年龄组体检正常儿童外周血淋巴细胞、多形核细胞及单核细胞3种白细胞的平均荧光强度(MFI)和阳性细胞百分率(PPC)。结果流式细胞检测表明,同年龄组内多形核细胞及单核细胞MFI和(或)PPC比较无显著性差异(P〉0.05),但2类细胞MFI和(或)PPC均显著高于淋巴细胞(Pa〈0.01);不同年龄组淋巴细胞、多形核细胞及单核细胞3种白细胞的MFI和(或)PPC比较有显著性差异(Pa〈0.01),其中以0-1岁组表达最高,2-3岁组次之,4-7岁组较低,8-15岁组表达最低。结论儿童外周血白细胞CAR表达以多形核细胞及单核细胞为主,流式细胞术是检测白细胞CAR表达的有效方法之一。

Abstract:Objective To investigate the expression of coxsackie-adenovirus receptor (CAR) on leukocytes of various types in peripheral blood in children.Methods The mean fluorescence intensity (MFI) and percentage of positive cell (PPC) in peripheral blood lymphocytes,monocytes and polymorphnuclear leukocytes of normal children were measured by flow cytometry.Results Flow cytometry analysis showed that the differences of CAR MFI and/or PPC between monocyte and polymorphnuclear leukocytes were not significant (P>0.05),but that MFI and/or PPC of CAR in the 2 types cells markedly increased compared with lymphocytes in the same group(Pa<0.01).The differences of CAR MFI and/or PPC among various groups were obvious(Pa<0.01).The expression levels of CAR were the highest in 0-1 year old group,the next were the 2-3 and 4-7 years old groups,and the lowest were those in 8-15 years old group.Conclusions The expression levels of CAR in peripheral blood leukocytes of children are higher than that in monocytes and polymorphnuclear leukocytes.Flow cytometry is a good technique to examine CAR expression.
