摘    要:目的:建立测定连翘中连翘苷含量的超高效液相色谱法。方法:色谱柱为Zorbax Eclipse PlusC1(8100mm×2.1mm,1.8μm),流动相为乙腈-水(24:76,V/V),流速为0.3mL·min-1,检测波长为277nm,柱温为室温。结果:连翘苷的进样量在5.9424~59.4240ng范围内与峰面积积分值呈良好的线性关系(r=0.9993);平均加样回收率为99.83%,RSD=1.27%(n=6)。结论:超高效液相色谱法与传统的高效液相色谱法相比,分析速度快、准确度高、重复性好,减少了有机溶剂的使用,可作为连翘药材质量控制的新方法。

Abstract:OBJECTIVE:To establish the UPLC method for the determine of phillyrin in Forsythia suspensa. METHODS:The determination was performed on a Zorbax Eclipse Plus C18(100 mm×2.1 mm,1.8 μm) column with mobile phase consisted of acetonitrile-water(24:76,V/V) at the flow rate of 0.3 mL·min-1. The detection wavelength was set at 277 nm and the column temperature was at the room temperature. RESULTS:The linear range was 5.942 4~59.424 0 ng(r=0.999 3) with an average recovery of 99.83%(RSD=1.27%,n=6) . CONCLUSION:UPLC method is rapid,accurate and reproducible,and it also use the less organic solvent than traditional HPLC method. The method can be used for the quality control of F. suspensa.
