玉竹初代培养最适条件的筛选

摘    要:以玉竹Polygonatum odoratum(Mill.)Druce]的叶片、茎段以及根茎作为外殖体,采用不同灭菌方式进行灭菌,在不同基本培养基上进行培养,添加不同种类生长调节剂,分析其最适的初代培养条件。结果表明,最适灭菌方法为75%乙醇处理10 s配合0.1%升汞处理7 min,其污染率为13.3%。根茎作为外殖体有较高的诱导率,达到90.0%;在培养中,最适宜初代培养的基本培养基为MS,其诱导率为86.7%;最适宜根茎分化的生长调节剂为MS+1.0 mg/L 6-BA+0.5 mg/L NAA,其次为MS+1.0 mg/L 6-BA+0.5 mg/L IAA,其分化率分别为80%和62%。

Abstract:Using Polygonatum odoratum (Mill.) Druce leaves,stem and rhizome as explants, and the explants were sterilized with different sterilization,incubated with different basic medium,in different types of additional hormone to investigate the op-timum primary culture conditions. The results showed that the optimum method of sterilizing time and 0.1% corrosive subli-mate 7 min with 75% alcohol 10 s,the pollution rate was 13.3%. Rhizome as explants had high induction rate,the induction rate was 90.0%. In culture,the most appropriate primary culture of basal medium was MS, the induction rate was 86.7%. Optimum hormone rhizome differentiation of 1.0 mg/L 6-BA+0.5 mg/L NAA,followed by 1.0 mg/L 6-BA+0.5 mg/L IAA,differ-entiation rate of 80% and 62%.
