多种维生素矿物质补充剂对小鼠抗氧化功能的研究

摘    要:目的 观察多种维生素矿物质补充剂的抗氧化作用。方法 颈背部皮下注射D-半乳糖建立小鼠衰老模型,同时分别灌胃给予不同剂量(0、0.09、0.18和0.36g/kbw)的受试物(多种维生素矿物质补充剂),试验期为60天。试验结束处死动物取血及肝脏测定血液及肝脏中丙二醛(MDA)、总抗氧化能力(T-AOC)和活性氧(ROS)水平及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,并测定血淋巴细胞DNA断裂情况。结果 与模型对照组比较,各受试物剂量组血浆及肝脏中MDA、ROS水平显著降低,除中剂量组肝脏SOD活性和低剂量组血浆和肝脏T-AOC外,其他各剂量组肝脏组织和血浆中SOD、T-AOC显著升高;血浆GSH-Px活性与彗星实验中的尾长和clive尾距在各组之间差异无显著性。结论 多种维生素矿物质补充剂具有一定抗氧化功能。

Abstract:Objective To study the antioxidative effect of multivitamin/mineral supplementation on mice. Methods Aging mouse model was established by subcutaneous injection of 8% D-galactose. These mice were simultaneously given multivitamin/mineral supplementation by gavaged at different doses(0, 0.09, 0.18, and 0.36 g/kg b w)for 60 days. Maleicdialdehyde(MDA), total anti-oxidative capacity(T-AOC), reactiveosygenspecies(ROS), superoxide dismutase(SOD), and glutathione peroxidase(GSH-Px)activities in blood and in liver were measured. In addition, lymphocytes were also separated for determination of DNA damage using comet assay. Results MDA level and ROS level in all multivitamin/mineral supplementation-treated groups were significantly lower than those in 0 g/kg group. SOD and T-AOC activities in part of multivitamin/mineral supplementation-treated groups were significantly higher than those in 0 g/kg group. GSH-Px activity and tail length and olive moment in comet assay had no significant difference among all multivitamin/mineral supplementation-treated groups and untreated group. Conclusions Multivitamin/mineral supplementation has the antioxidative function.
