瑞舒伐他汀对扩张型心肌病大鼠心肌组织赖氨酰氧化酶表达水平的影响

摘    要:目的:探讨瑞舒伐他汀对扩张型心肌病大鼠心肌组织赖氨酰氧化酶(LOX)表达的影响。方法成年大鼠50只,随机分为三组:对照组(NS 组:n=10只,腹腔注射等量生理盐水);扩张型心肌病模型组(DCM 组:n=20,腹腔注射阿霉素);干预组(GY 组:n=20,腹腔注射阿霉素,同时给予瑞舒伐他汀灌胃)。10周末处死大鼠,HE、Masson 染色测定心肌组织胶原纤维化情况;提取左室心肌组织 RNA 及蛋白质,RT-PCR 法及 Western blot 测定心肌组织 LOX 的 mRNA 及蛋白表达水平。结果 HE、Masson 染色显示 DCM 组大鼠心肌组织明显纤维化,而 GY 组大鼠心肌组织纤维化程度较轻;DCM 组大鼠心肌组织 LOX 的 mRNA 及蛋白表达水平明显高于 NS 组(P <0.01);与 DCM 组相比,GY 组心肌组织 LOX 的 mRNA 及蛋白的表达明显下调(P <0.01)。结论瑞舒伐他汀可逆转DCM 大鼠心肌组织纤维化及 LOX 的表达。

Effects of rosuvastatin on expression of lysyl oxidase in rat heart with dilated cardiomyopathy
Abstract:Objective To explore the effects of rosurvastatin on expression of lysyl oxidase (LOX)in myocardial tissue of rats with dilated cardiomyopathy (DCM).Methods Fifty SD rats were randomly divided into 3 groups which were control group (n=10,equivalent normal saline was intraperitoneal injected),DCM group (n=20,2.5 mg· kg-1 adriamycin was intraperitoneal injected every time,one time every week for 6weeks),and treatment group (n=20,2.5mg· kg-1 adriamycin was intraperitoneal injected every time,one time every week for 6weeks,gastric lavage in rosurvastain 3 mg·kg-1 ·d-1 at the same time).At the end of week 10,mice were sacrificed,HE and Masson pathological staining,analysis of degree fibering in the myocardial tissue,expression level of LOX mRNA and protein in myocardial tissue of DCM rats were measured by RT-PCR and western blot (WB).Results HE and Masson pathological staining was shown myocardial tissue significantly fibering in DCM group than that in control group,and significantly decreased in treatment group.The expression level of LOX mRNAs and protein were sig-nificantly increased in DCM group than that in control group (P <0.01),and were significantly decreased in treat-ment group than that in DCM group (P <0.01).Conclusion rosurvastatin can attenuate the increased-expression of LOX and fibering in myocardial tissue of DCM rat.
