过表达NPM1对胰腺癌细胞增殖、凋亡影响的研究*

摘    要:目的:探讨NPM1过表达对胰腺癌细胞增殖、凋亡的影响。方法:对人胰腺癌细胞株AsPc-1,BxPc-3,PANC-1,CFPAC-1及人正常胰腺细胞株HPDE6CT作传代培养,检测其中NPM1表达。选择表达较低的细胞行NPM1过表达质粒转染,并设立空白质粒转染对照组。48小时后,采用RT-PCR及Western检测NPM1表达水平变化,并行MTT及流式细胞技术检测细胞增殖、凋亡情况。结果:(1)NPM1在BxPc-3细胞系中表达最低;(2)NPM1过表达组与空白质粒组相比,NPM1的mRNA表达量及蛋白表达量明显增多,差异显著(P<0.05);(3)NPM1过表达组相比空白质粒组,细胞增殖增加,早期凋亡细胞明显减少(P<0.05)。结论:NPM1的过表达可促进胰腺癌细胞增殖,减少凋亡。

Abstract:Objective: To discuss the proliferation and apoptosis of pancreatic cancer cells by over expressing NPM1. Methods:Human pancreatic cancer cell lines AsPc-1,BxPc-3,PANC-1,CFPAC-1 and human normal pancreas cell lines HPDE6CT were chosen to be cultured. Then the cells which had lower expression of NPM1 were chosen to transfer NPM1-overpression plasmid and the control groups were set. 48h after transfection, RT-PCR and Western Blot were used to test the expression of NPM1. MTT and flow cytometry were adopted to test the proliferation and apoptosis of cells. Results:(1)NPM1 was least expressed in BxPc-3 cells. (2)Compared to the blank plasmid group, the NPM1 overexpression group had higher mRNA and protein expression of NPM1(P<0.05).(3)The results of MTT and flow cytometry: compared to the NPM1-control group, cells in the NPM1-overexpression had higher proliferative capacity and less apoptosis(P<0.05).Conclusions: The increase of NPM1 could improve the proliferative capacity and decrease the apoptosis of pancreatic cancer cells.
