摘    要:目的检测体外培养的人神经干细胞端粒酶活性,分析其与神经干细胞生物学特性的关系。方法分离、鉴定并扩增取自10~14周药物流产胎儿大脑皮质的神经干细胞,应用PCR-ELISA法检测神经干细胞端粒酶活性。结果经免疫组化检查,培养的细胞为人神经干细胞;其在体外培养过程中,仅在早期10代表达极低的端粒酶活性(相对活性为6.4%~21.2%),其后消失。结论神经干细胞表达端粒酶活性,但较低且不能长期维持,这可能是神经干细胞不能长期传代的原因之一。

Abstract:Objective To detect the telomerase activity in human neural stem cells, and analyze the relation between telomerase activity and the biological characteristics of human neural stem cells. Methods The neural stem cells were isolated, identified and amplified from the cortex of aborted human embryo with 10 to 14-week gestational age, then telomerase activity was detected by PCR-ELISA. Results Immunohistochemical test showed that the cultured cells were neural stem cells. At early 10 passage human cultured neural stem cells expressed very low levels of telomerase with the relative activity of 6.4% to 21.2%, and disappeared whereafter. Conclusion Human neural stem cells express low level of telomerase activity, and could not maintain for long time. This may be the one of the reasons that human neural stem cells could not be long-term passaged in vitro.
