摘    要:目的: 运用生物信息学技术分析胃癌中microRNA表达特征。方法: 使用R语言软件包分析GEO芯片数据集的两个子集中的miRNA表达量的差异;采用qRT-PCR技术检测本院60例胃癌患者癌组织及20例胃癌手术患者癌旁组织中miR-125b-5p的表达量,统计分析其表达特征与分化程度、TNM分期、性别、年龄、家族史及肿瘤直径之间的关系。结果: miR-125b-5p在GSE93415和GSE99415中的表达均上调,且其在胃癌组织与癌旁胃组织表达量差异具有统计学意义(P<0.05)。使用qRT-PCR验证了miR-125b-5p在胃癌组织中的表达高于癌旁组织的趋势,miR-125b-5p表达量与TNM分期和浸润程度有关(P<0.05),与年龄、性别、家族史、肿瘤直径和分化程度均无关(P>0.05);高表达miR-125b-5p的胃癌患者生存期明显缩短。结论: 胃癌组织miR-125b-5p高表达,且与胃癌TNM分期、浸润程度及总生存率有关。

Abstract:Objective: To analyze the expression of microRNA in gastric cancer by bioinformatics. Methods: The differential expression of microRNAs in two subsets of GEO dataset was analyzed. Using R software package. The expressions of microRNAs in 60 gastric cancer surgical specimens and 20 paracancerous surgical specimens were detected by qRT-PCR. The relationships between microRNA expression and differentiation, TNM stage, sex, age, invasion, family history and tumor diameter were analyzed. Results: The expression of miRNA-125b-5p was up-regulated in GSE93415 and GSE99415, and the difference was statistically significant(P<0.05). The similar expression trend was confirmed in gastric cancer and normal gastric tissues with qRT-PCR. The expression of miRNA-125b-5p was correlated with TNM stage and invasion degree(P<0.05), but not with age, sex, family history, tumor diameter and differentiation degree(P>0.05).The survival time of gastric cancer patients with high expression of miR-125b-5p was significantly reduced. Conclusion: miR-125b-5p is associated with TNM stage, invasion and survival time of gastric cancer. [Key words]miR-125b-5p; gastric cancer; bioinformatics; GEO microarray datasets
