HPLC法同时测定泌感颗粒中盐酸小檗碱和槲皮素的含量

目的建立同时测定泌感颗粒中盐酸小檗碱和槲皮素含量的方法。方法采用HPLC法。用Diamonsil C18柱(200 mm×4.6 mm,5μm)分离,以乙腈-质量分数3%的磷酸溶液(体积比34∶120,用三乙胺调pH至3.0)等度洗脱,流速为1.0 mL.min-1,柱温为30℃,检测波长为360 nm。结果盐酸小檗碱质量浓度在1.04~10.4 mg.L-1内、槲皮素质量浓度在0.400~4.00 mg.L-1内与峰面积呈良好的线性关系,r分别为0.999 9和0.999 8;平均回收率分别为98.5%(RSD=0.92%)和98.8%(RSD=1.43%)。结论HPLC法可作为泌感颗粒的质量控制方法之一。

Simultaneous determination of contents of berberine hydrochloride and quercetin in Migan granules by HPLC

To establish an HPLC method for simultaneous determination of berberine hydrochloride and quercetin in Migan (traditional Chinese medicines) granules. Methods The chromatographic method was carried out on a Diamonsil C18 column (200 mm × 4.6 mm, 5 μm) with acetonitrile-3 % phosphoric acid solution (V:V =34:120， adjust pH value to 3 by triethylamine) as the mobile phase. The flow rate was 1.0 mL· min-1 and the column temperature was 30 ℃. The detection wavelength was set at 360 nm. Results The calibration curve was linear within the range of 1.040-10.40 mg·L-1 (r=0.999 9) for berberine hydrochloride and 0.400 -4.00mg·L-1 (r=0.999 8) for quercetin, the average recoveries were 98.5 % (RSD=0.92 %) for berberine hydrochloride and 98.8 % (RSD=1.43 %) for quercetin. Conclusions The method is simple, accurate and reproducible, which can be used for the quality control of Migan granules.
