| 摘 要： | 目的研究在体外Claudin-1(CLDN1)单克隆抗体对丙型肝炎病毒(hepatitis Cvirus,HCV)感染的阻断效应。方法应用基因免疫接种制备CLDN1单克隆抗体,研究该抗体在HCV细胞感染模型(HCV cell culture,HCVcc)中的阻断效应。结果 CLDN1单克隆抗体和人源性CLDN1受体高亲和性结合,且对肝肿瘤细胞系(Huh7.5.1)无细胞毒性。在HCV体外感染模型中,CLDN1单克隆抗体可剂量依赖性抑制HCV感染,且对HCV主要基因型(1型及2型)均有效。结论 CLDN1单克隆抗体有望作为一个新的抗病毒手段应用于HCV感染的防治。
