| 摘 要： | 在利用基因工程方法实现大肠杆菌表达外源重组人ＶＥＧＦ的基础上，应用制备性ＳＤＳ－ＰＡＧＥ从包涵体中分离纯化了这一因子。纯化蛋白在含有氧化型与还原型谷胱甘肽和肝素的缓冲液中复性，复性后的ＶＥＧＦ蛋白成为同源双体，经Ｍｉｌｅｓ方法检测，证明可提高血管通透性，具有生物活性。这一研究为制备具有生物活性的重组人ＶＥＧＦ蛋白，从而开展有关其生理与病理意义的研究奠定了基础。

| Abstract: | On the basis of expressing recombinant human VEGF in E. coli we reported earlier, in this study wereport the isolation and bioactivity recovery of this factor from the procaryotic expression products. Isolatedrecombinant human VEGF is renatured in buffer containing oxidizedand reduced glutathione and heparin,the renatured VEGF forms homodimer shown in non-reduced analytical SDS-PAGE. Miles assay shows therenatured VEGF promotes vessel permeability in dose-potency manner. |
