苦参碱对宫颈癌HeLa细胞的作用

目的:观察不同浓度的苦参碱对宫颈癌HeLa细胞的作用。方法:浓度分别为0.5,1.0,1.5,2.0 g/L的苦参碱体外培养HeLa细胞24,48,72 h后,分别用MTT法观察苦参碱对HeLa细胞的生长抑制作用、AnnexinV-PI双标染色用流式细胞仪检测苦参碱对HeLa细胞凋亡的影响。结果:不同浓度(0.5,1.0,1.5,2.0 g/L)的苦参碱具有抑制宫颈癌HeLa细胞增殖的作用,呈明显的时间、剂量依赖性。各浓度和时间之间比较具有显著差异性(P<0.01)。FCM检测结果显示苦参碱作用后,细胞的凋亡率增加,呈现随时间和剂量依赖性。各浓度和时间之间比较也具有显著差异性(P<0.01)。结论:苦参碱对人宫颈癌HeLa细胞的增殖具有明显的抑制作用,能诱导细胞凋亡的凋亡。

Effect of Matrine on Cervical Cancer HeLa Cells

Objective:To study the effect of Matrine at different concentrations on cervical cancer cells.Methods:Matrine at different concentrations(0.5,1.0,1.5 and 2.0 g/L) was respectively used to cultivate cervical cancer HeLa cells in vitro.After HeLa cells were cultivated for 24,48 and 72 hours respectively,the effect of Matrine on cervical cancer cells was assayed by MTT,flowcytometry(FCM) and RT-PCR method respectiveLy.Results:Matrine inhibited the proliferation and induced apoptosis of HeLa cells on dose-and time-depending manner,and the differences among different concentration and time groups were significant(all P<0.01).The expression of survivin mRNA was also decreased.Conclusion:Matrine restrained the proliferation of HeLa cells significantLy,and the decreasing of survivin Marine may be one of the mechanisms of cervical cancer HeLa cell apoptosis induced by Matrine.
